| Objective:To study the different interventions on hypertension related vascular dementia rats’ differences of blood pressure, praxiology and brain function beforeand after the therapy, and provide new ideas and methods for the treatment of vascular dementia.Methods:The improved2-VO (2-VO) and high salt diet were used to establish rat models. The rat tail artery blood pressure measuring instrument and the Morris water maze was used to screen hypertension related vascular dementia model rats. The model rats were divided into4groups:the model group, electro-acupuncture group, point application group,Electro-acupuncture and acupoint application treament were given respectively. Artery blood pressure measuring instrument for measuring each rat’s blood pressure, Morris water maze test the ability of learning and memory before and after the treatment. Functional magnetic resonance imaging (fMRI) technology and fractional amplitude of low frequency fluctuation(fALFF) calculations were used to detect the differences in different brain regions.Results:1. Blood PressureCompared with the model group, after treatment in the treatment groups have significantly lower blood pressure (P<0.05), with the extension of treatment time, the effect on blood pressure of rats become more and more obvious; Blood pressure is influenced by time factor, the difference was statistically significant at different time point (P<0.05); Interaction was existed between different groups and times on blood pressure (P<0.05), it means the rats’blood pressure changes with time have a certain tendency.2. Learning and memoryPlace navigation showed:Compared with the model group,the EA group has significantly shorter escapelatency, and with statistical significance (P<0.05); no statistical significance (P>0.05)was showed on the escapelatency between the point application group and model group.spatial probetest showed:Compared with the model group,the number of cross-platform was significantly increased on the EA group, and with statistical significance (P<0.01); no statistical significance (P>0.05)was showed on the number of cross-platform between the point application group and model group. 3. Functional magnetic resonance imagingAfter EA treatment, the ALFF value increased in PC,CA1-CA3,M1,M2and cg region, and reduced in fusiform gyrus, amygdala ang PPC region; after the point application treatment,the ALFF value increased in cg, PRh, Ect and cc region,and reduced in the S2region.Conclusion:1.The treatment of electroacupuncture and point application can effectively reduce the blood preesure of rats.and the antihypertensive effect is more significant with the prolonged treatment.2. After the treatment of electroacupuncture,the ability of learning and memory is significantly improved,but after a course of point application treatment,the learning and memory ability of rats had no obvious promotion.3. After EA treatment, there were multiple ALFF changes in the brain regions related to cognitive function, and the application therapy lead to the ALFF changes in olfatory related brain regions that indirectly related to cognitive function.4. After different treatments, the ALFF value has differences in different r-egions of the rat, fMRI have important reference value to evaluate the effects of different therapy on VD brain function. |
