| Papillary thyroid carcinoma is the most common thyroid carcinoma,and is also the most common endocrine malignancy. The incidence ofpapillary thyroid cancer in China increases significantly in recent years.The papillary thyroid carcinoma-associated protein and gene couldfacilitate the diagnosis of papillary thyroid carcinoma and help determinethe degree of malignancy, rendering it a hotspot for researches. Studieshave shown that kiss-1, ki-67and VEGF-C discriminate normal thyroidtissue and thyroid carcinoma, indicating that they might be used in theauxiliary diagnosis of PTC. The IHC was used to analyze the expression ofkiss-1, ki-67and VEGF-C in PTC and follicular adenoma (FA), andmicroscopic image analysis technique was used to analysis the differences.The study tries to explore:1. The expression of kiss-1, ki-67andVEGF-C in PTC and FA respectively, and their relationship between theexpression of the three indicators and the general conditions, thepathological features, i.e., lymph node metastasis and capsular invasion;2.The interrelation expression of kiss-1, ki-67and VEGF-C in PTC and FA,respectively;3. If it is possible to employ microscopic image analysistechnique in the detection of thyroid cancer tissue-specific proteinexpression.All paraffin-embedded blocks of PTC and FA used in the presentstudy were retrieved during a year period (2011-2012) from archives ofthe Department of Pathology in the PLA309hospital. Paraffin-embeddedtissues from44cases of PTC were used, while the tissues of12cases of FAwere used as a control. Immunohistochemical staining was performed on kiss-1, ki-67and VEGF-C respectively, and microscopic image analysistechnique was used to analyze the staining results of kiss-1, ki-67andVEGF-C.It was found that:1. The IOD and positive rate of kiss-1in the PTCgroups were (475.56±126.02,40.90%), respectively. It was significantlylower than that in FA(590.30±166.19ã€75.00%);2. The IOD and positiverate of VEGF-C in the PTC groups were (805.29±226.05,68.18%),respectively. It was significantly higher than that in FA(630.76±169.61,33.33%);3. The proliferation index and positive rate of ki-67in the PTCgroups were (3.36±1.11)%,61.36%, respectively. It was significantly lowerthan that in FA(1.89±0.68)%,25%;2. The expression of kiss-1in lymphnode metastasis group and capsular infiltration group was significantly lessthan in the group without lymph node metastasis and group withoutcapsular infiltration(P<0.05). The more expression of kiss-1was, the lessTNM staging was(P>0.05), however, there had no significant relationshipwith patient’s age and sex(P>0.05). The expression of VEGF-C in lymphnode metastasis group and capsular infiltration group was significantlymore than in the group without lymph node metastasis and group withoutcapsular infiltration(P<0.05). The more expression of VEGF-C was, themore TNM staging was(P<0.05), however, there had no significantrelationship with patient’s age and sex(P>0.05). The expression of ki-67inlymph node metastasis group and capsular infiltration group wassignificantly more than in the group without lymph node metastasis andgroup without capsular infiltration(P<0.05). The more expression of ki-67was, the more TNM staging was(P<0.05), however, there had nosignificant relationship with patient’s age and gender(P>0.05).3. Weanalyze the expression of kiss-1, ki-67and VEGF-C in forty-four PTC andtwelve FA tissues by using Pearson correlation analysis. It showed thatkiss-1and ki-67, VEGF-C had negative correlation with PTC and FAtissues, while ki-67and VEGF-C had positive correlation with PTC and FA tissues.According to our study, we conclud that kiss-1, ki-67and VEGF-Cmay play an important role in PTC.1. Involved in tumor occurrence; theproliferation index and positive rate of ki-67in PTC is higher than and inFA.2. Involved in tumor invasion and metastasis; kiss-1and VEGF-C canbe used to measure the invasion and metastasis of PTC;3. Linearcorrelation analysis shows that kiss-1is negative correlated with ki-67,VEGF-C in PTC and FA tissues, while ki-67is positive correlated withVEGF-C in PTC and FA tissues, indicating kiss-1, ki-67and VEGF-C playan important role in cell proliferation, invasion and metastasis;4.Microscopic image analysis can analyze the result of IHC staining byquantifying the difference, and it is an effective method for the detection ofthyroid tumor marker, facilitating the diagnosis of PTC and predicting theprognosis of the tumor. |
