Objective:To discuss the feasibility of8-OHdG which is used as a biomarker for diagnosing breast cancer by detecting the urinary8-hydroxydeoxyguanosine (8-OHdG) levels in trial groups.Methods:Collecting50cases of female patients with breast cancer in department of hepatobiliary surgery, People’s Hospital of Guangxi Zhuang Autonomous Region, during December2012to July2013. All of them were treated by breast cancer radical resection and underwent chemical treatment after operation. As while, collecting50cases of benign breast tumor which include fibroadenoma of breast and hyperplasia of breast.All the patients were confirmed by pathologic examination and none of them with serious lesions of vital organs such as heart, lung and kidney disease and drug abuse history Collecting50cases healthy women of the physical examination center as the control group. Urinary8-OHdG levels in different periods of breast cancer, benign breast tumor and control groups were detected by ELISA and serum CA15-3、 CA12-5、CA19-9、CEA、AFP were detected by radioimmunoassay.Results:It was found that the excretion levels of urinary8-0HdG in preoperative breast benign tumor patients had no significant difference to healthy adults [(16.60±6.83) vs (17.77±4.3) ng/mgCr,P>0.05]. Also, no significantly difference was found between preoperative and postoperative breast benign tumor patients. On the other side, urinary8-OHdG levels of preoperative breast cancer patients were significantly higher than healthy adults [(23.34±5.3)vs(17.77±4.3) ng/mgCr], P<0.05]. Patients receiving modified radical mastectomy showed a significant decrease in urinary8-OHdG levels [(23.34±5.3)vs(18.55±4.44) ng/mgCr, P<0.05], and closed to the levels of healthy adults. There was a remarkable increase of urinary8-OHdG from the patients receiving chemotherapy after one week [((31.34±4.32) vs (18.55±4.44) ng/mgCr]. However, after a month following surgery the patients showed a decrease of the excretion levels and closed to the levels before chemotherapy [(19.72±4.77) ng/mgCr].Conclusion:1) There is a relationship between oxidative DNA damage and breast cancer.There weren’t correlations among CA15-3、CA12-5、 CA19-9、CEA、AFP and8-OHdG;2)Urinary8-OHdG levels might be an useful biomarker for diagnosing breast cancer and evaluating the effect of surgical therapy;3)Chemotherapy increased oxidative DNA damage,whether it can evaluate the effect of chemotherapy or not,which need more cycles to trace;4) Expression of8-OHdG was significantly lower in triple-negative breast cancer (TNBC)than in the non-TNBC group,which may be promising targets when elucidating the pathogenesis of TNBC.
