Objective:To study the radiosensitization effect and cytotoxicity of Sodium Danshensu on Lewis Lung Carcinoma cell in vitro, with the application of cell colony formation and MTT.Methods:Culture the Lewis Lung Carcinoma cell,and the exponentially growing cells at a cell density of6×103cells/well were inoculated into96-well plates to give sodium Danshensu at a certain concentration gradient, research the cytotoxicity of sodium Danshensu of Lewis lung carcinoma cells by MTT assay,so calculated the IC50, IC10values. IC10values conduct the working concentration of radiosensitization. At the same time using the same MTT method calculate the IC50, IC10values of cisplatin on Lewis lung carcinoma cells.Research the Danshensu sodium from the body of Lewis lung carcinoma cells in vitro radiosensitizing effect using the cell colony formation method. The experiment was divided into normal control group, the radiation alone group, simple drug groups, drug and radiation group, positive control group, and the positive control group were treated with cisplatin.Lewis lung carcinoma cells in the culture flasks were added to a concentration of IC10values Danshensu sodium and cisplatin before irradiation, which were given0Gy,0.5Gy,1Gy,2Gy,3Gy,4Gy,6Gy,8Gy,10Gy irradiation dose gradient. Cells in the culture flasks after irradiation was placed in0.5%CO2thermostat at37℃for4hours,and discarded original culture medium, digestion, counting. The number of the cells to the dish planting determined according to the cell culture flask received different doses of irradiation,and each of three petri dishes. The dishes were cultured for14days in a37℃,0.5%CO2thermostat. Count of the colony forming for calculating the survival fraction and colony formation rate.The mathematical model of the single-hit multi-target was used to plot survival curve.Calculate N, Do,Dq,SERDO,SERDq,SERSF2, The strength of radiosensitizing effect of each group were compared.Results:The IC50value of Danshensu sodium on the Lewis lung carcinoma cells was77.75ug/ml,the IC10value was14.39ug/ml.The IC50value of Cisplatin on the Lewis lung carcinoma cells was51.35ug/ml,the IC10value was11.18ug/ml. Lewis lung carcinoma cell colony formation rate was66%.The values of N,D0,Dq of Lewis lung carcinoma cells received treatment of radiation alone were1.975,1.5475Gy,1.5354Gy; the values of N, Do,Dq,SERDo,SERDq,SERsF2of Lewis lung carcinoma cells received treatment of radiation+Danshensu sodium were1.635,1.3639Gy,0.8661Gy,1.1346Gy,1.7728Gy,1.4269Gy; the values of N, Do,Dq,SERDo,SERDq,SERSF2of Lewis lung carcinoma cells received treatment of radiation+Cisplantin were1.492,1.2697Gy,0.6247Gy,1.2188Gy,2.4578Gy,1.7759Gy.Conclusions:The result of in vitro experiments has shown that the sodium Danshensu has the cytotoxicity and radiosensitizing effect at the IC10concentrations of Lewis lung carcinoma cells,but which was not better than Cisplantin.
