Objectives: To explore the role of Beclin1, LC3in hyperoxia-induced injury of alveolar epithelial cells A549.Methods: A549cells were cultured in vitro and divided randomlyinto control (incubated with5%CO2), hyperoxia group (exposed to amixture of900ml/L O2and50ml/L CO2at speed of3L/min for10mins, then cultured in a closed environmen).The cellularmorphologywas observed under inverted microscope at12,24and48h of treatment.Expression of Beclin1,LC3were detected by immunohistochemistryafter12h,24h,48h of treatment.Results: Compared with the control group, autophagy-relatedproteins Beclin1, LC3have lower levels of expression in the lung cell inhyperoxia group, and the expression levels of Beclin1,LC3showed atime-dependent increase in24hours, and reached the highest in48hours (all P <0.05).Conclusions: Beclin1, LC3could play an important role inmaintaining cell homeostasis and cell protective effect in the process ofoxidative stress injury induced by hyperoxia.
