Objective:Explore the effect of Mycobacterium tuberculosis PhoPR two-component system overexpression on drug resistance of rifampicin-resistant strain of Mycobacterium tuberculosis clinical isolates in Xinjiang.Methods:Construction and identification the recombinant shuttle plasmid pMV361-PhoP, pMV361-PhoR and pMV361-PhoPR of Mycobacterium tuberculosis PhoPR two-component system, then introduced into a rifampicin-resistant strain of Mycobacterium tuberculosis clinical isolates in Xinjiang by electroporation. Using real-time quantitative PCR technology to detect and identify the over-expression of PhoP, PhoR and PhoPR gene in rifampicin-resistance of Mycobacterium tuberculosis clinical isolates in Xinjiang was successfully constructed. Determination of minimal inhibitory concentration by using colorimetric method to detect and investigate the resistance changes of over-expression of PhoP, PhoR and PhoPR gene in rifampicin-resistance of Mycobacterium tuberculosis clinical isolates in Xinjiang.Results:Construction and identification the recombinant shuttle plasmid pMV361-PhoP, pMV361-PhoR and pMV361-PhoPR. The results showed that over-expression of PhoP, PhoR and PhoPR gene in rifampicin-resistance of Mycobacterium tuberculosis clinical isolates in Xinjiang can significantly increased the resistance.Conclusions:The over-expression of PhoP, PhoR and PhoPR gene can significantly increased the resistance of rifampicin-resistant strain of Mycobacterium tuberculosis clinical isolates in Xinjiang, it could be used for the further study of Mycobacterium tuberculosis PhoPR two-component systemon mechanism on drug resistance of Mycobacterium tuberculosis clinical isolates in Xinjiang.
