Objective:The experiment aims to explore and study the regulatory influencing mechanism of mycobacteriumtuberculosis on iron metabolism of infected host macrophages through establishing the model of micealveolar macrophages infected by mycobacterium tuberculosis.Methods:The Laboratory Mice were infected respectively bacterial suspension of the international standard virulentstrain of Mycobacterium tuberculosis H37Rv strains(H37Rv), Bacillus Calmette-Guérin strains (BCG) andsterile saline solution(normal control) by tail vein. After the successful replication of each group of miceinfection model, we cleaved the alveolus of every group of mice and collected lavage fluid of mice toobtain alveolar macrophages of the infected mice on the1st,3rd,5th,7th,9th,11th,13th,15th day, thendetecting protein expression of the Fn、TfR、Lf、FPN in macrophage culture supernatants of the threegroups and analyzing the1st,3rd,5th,7th,9th,11th,13th,15th day differences by ELISA in each time phase;Using Western-Blot to make comparative research on protein expression of Fn and TfR in different timephase.Results:After being infected by the strains, the expression of Fn in macrophages decreased with time going; and atthe time point of7th,9th,11th, the difference had statistical significance (P<0.05).The expression of TfR in mice alveolar macrophages gradually increased after the infecte the mice, which7th,9th,11th were highest in the Infection and H37Rv group was higher than the expression of BCG group,the difference was statistically significant (P <0.05).The expression of FPN were gradually decreased after infecte the mice,and5th,7th,9th were the mostobvious than other days, the difference was statistically significant (P <0.05).The strains might induce mice alveolar macrophages to secrete higher expression level of Lf after beinginfected. The expression of Lf at time points of7th,9th,11th were increased the most obvious than those innormal control group, all of which showed statistical significance (P<0.05).Conclusions:The decreased of Fn in the alveolar macrophages of mice, suggesting that infection leads Fe3+to decreased.The increased of TfR and Lf alveolar macrophages of mice macrophages, suggesting that activation of thebinding iron uptake pathway.The decreased of FPN in the alveolar macrophages of mice, suggesting that mediated iron transferring-outpathway...
