Objective: To explore the clinicopathological and prognosis significance as well as the impact ofconstitutively activated Stat3in gastric cancer through detecting SOCS1and SOCS3expression incancerous and adjacent non-cancerous tissues.Methods:(1) Using specific antibodies, the levels of SOCS1and SOCS3were tested in the formalin-fixedand paraffin embedding tissues, including118cases of gastric cancer and adjacent tissues57cases (normalgastric mucosa29cases and intraepithelial neoplasia28cases) by Immunohistochemistry (Envisionmethod).(2) All clinicalpathology and follow-up data of the118patients were collected.(3) Statisticalanalysis was using SPSS (version17.0) statistical software package, Qualitative data were analyzed usingchi-square test and paired-samples Wilcoxon test. Spearman rank correlation test was used to analyze theassociation between SOCS1, SOCS3expression and Stat3activation in different tissues. The relationshipbetween survival rates and SOCS1, SOCS3were calculated by the Kaplan-Meier curve and the log-ranktest.Results:(1) The expressions of SOCS1and SOCS3in gastric carcinoma tissues are significantly weakerthan those in adjacent tissues (P<0.01), suggesting SOCS1and SOCS3may play an important role in thecarcinogenesis process.(2) The expressions of SOCS3become significantly weaker in pace with increasedmalignant behavior, such as the depth of invasion, lymph node metastasis and clinical stage, indicatingSOCS3may take part in the progression of gastric cancer.(3) The connection between SOCS3(notSOCS1) expression level and survival time of patients in gastric cancer is statistically significant (P=0.001), indicating that abnormal expression of SOCS3is a strong indicator to determine the prognosis ofpatients with gastric cancer.(4) The level of SOCS1, SOCS3expression and p-Stat3activation aresignificantly negatively correlated in GC (r=-0.198,-0.415). Decreased SOCS1, SOCS3might beimportant factors for p-Stat3activation gradually increased, suggesting diminished expression levels ofSOCS1, SOCS3involve in development of GC via up-regulating activation levels of Stat3.Conclusions:(1) Decreased SOCS1can up-regulating Stat3activation level, which may act as a key role inthe development of gastric cancer.(2) The low levels of SOCS3are correlated with development,progression and poor survival via regulating the activation of Stat3, suggesting SOCS3may be served as apotential predictor for the diagnosis, treatment and prognosis of gastric cancer.
