ObjectiveTo explore mRNA significance and expression of Notch1in clear cell renal cellcarcinoma(ccRCC) and the relation with clinical pathology factors. Furthermore,to explorethe Notch’s expression in renal carcinoma and silence the expression of Notch1by usingsiRNA and watch its effect about the proliferation of Caki-1.MethodsCollected52cases of renal carcinoma and30cases of normal kidney tissue specimens.The levels of Notch1expression was tested by RT-PCR and immunohistochemistry inpathologically identified ccRCC and normal kidney tissue. Notch1and clinicopathologicalparameters analyzed the correlation. Normal human renal tubular epithelial cell and renalcarcinoma lines were cultured in vitro. To detect expression of Notch1mRNA by real-timequantitative and used Western-blot Notch1protein levels, siRNA interference changed intoRCC Caki-1cells, transfection with Lip2000about48hours, By RT-PCR and Westernblot were checked among Notch1mRNA and protein expression. The cell of proliferationchanges was used CCk-8and Colony formation.ResultsThe masculine staining rates of Notch1in ccRCC was taller than those in normalrenal cell tissues (94.2%vs40.0%, P <0.05). Notch1level of expression is related withthe distant metastasis of kidney carcinoma, attacks higher than no-metastatic(P<0.05);Also relationship with tumor volume, the tumors≥7cm in diameter higher than thediameter <7cm tumors (P <0.05). Notch1in kidney cell carcinoma with high expressionrelatively with Normal kidney tubular epithelial cells, which has the differention wasstatistically significant (P <0.05). After specific siRNA interference can significantlyreduce the Caki-1cells, mRNA and protein expression, statistically dissimilarity (P <0.05),and Caki-1cells proliferation was obversious decreased, and difference was significantly(P <0.05). ConclusionNotch1is above expressed in the renal and kidney cell carcinoma may developrelevant. Meanwhile, Notch1and clinical staging of renal carcinoma, classification andtumor volume were related, with the diagnosis and therapy of kidney cell carcinoma cancerhave a certain significance. Notch1expression in renal clear cell carcinoma than in normalkidney cells, siRNA interference after Notch1can inhibit renal cell carcinoma Caki-1proliferation, adjusting the growth of kidney cancer, diagnosis and treatment for kidneycell carcinoma cancer have a certain significance.
