Objective:To study the expressions of cancerous inhibitor ofPP2A(CIP2A) and Death-associated protein kinase(DAPK) in different cervicaltissues and elucidate their significance in the initiation and development ofcervical cancer.Methods:1.38cases of cervical cancer tissues11cases ofcervical intraepithelial neoplasia(CIN)and10cases of normal cervical tissuesfrom patients were collected from the obstetrics and gynecologic department inthe Affiliated Hospital of Luzhou Medical College since2012October to2013October. The average age of patients in the three groups were44.80±12.57years old,48.82±11.05years old,45.47±7.31years old in normal cervixgroups, CIN groups and cervical cancer groupsrespectively.2.Immunohistochemical Envision method was used to detect theexpressions of CIP2A and DAPK in these cervical tissues.3.ImageproPlus5.0software was used to analyze the mean optical density(MOD) in theexperimental picture, then independent sample t-test was applied for thestatistics. Spearman rank correlation analysis was conducted for the correlationanalysis and P <0.05was treated as statistically significant in two-sidedtest.Results:1.CIP2A protein was mainly expressed in the cell membraneand(or) the cytoplasm in different cervical tissues. The MOD values ofCIP2A were10.27±18.19,46.71±39.23,61.96±39.06in normal cervix, CIN, cervical cancer tissues respectively. There was an increasing expressiontrend with the rise of disease severity. There was a significant expressingdifference of CIP2A between NCE and CC groups (P<0.001), NCE and CCgroups. But there was no significant difference between CIN and CCgroups(P>0.05). There was a significant difference in the different depth ofinvasion groups (P <0.05),while the other clinical pathological indexes wasnot(P>0.05).2. DAPK protein was positively expressed in the cell nucleusand(or) the cytoplasm in NCE, while the positive expression of CIN and CCtissues was located in the cell membrane and(or) the cytoplasm. The positivereaction showed different degrees of yellow or brown. The MOD values werenot obeyed normal distribution after K-S normal test, then transformed the databy square root so as to be applied. After transformation, the MOD values ofDAPK were9.04±3.47、9.04±3.47、3.74±3.30in NCE,CIN and CC respectively.There was a decreasing expression trend with the rise of disease severity.There was no significant expressing difference of DAPK between NCE andCIN groups (P>0.05),while there was significant expression difference ofDAPK between NCE and CC groups, CIN and CC groups(P<0.05). Therewas a significant difference in tumor maximum diameter between the≥4centimeters groups and<4centimeters groups (P=0.015<0.05). It was foundthat there was significant DAPK expression difference between the age of≥35years groups and <35years groups which tested by t-test with the limit of35years old (P=0.026<0.05). But there was no significantly difference in theother clinical pathological indexes (P>0.05).3There was no correlation between CIP2A and DAPK expressed in cervical tissues (P>0.05).Conclusion:1.CIP2A was overexpressed in cervical cancer while the expression of DAPKexpressed missing or reducing in cervical cancer.2. The increased expression ofCIP2A and the decreased expression of DAPK may be associated with theoccurrence of cervical cancer.3.There was not relationship betweenthe expression of CIP2A and DAPK in cervical cancer tissues which mayindicate the different pathway affecting the occurrence and development ofcervical cancer.
