Objective: Rat model of POCD was established by anesthetic isoflurane undergoingsplenectomy, explore the pathogenesis mechanism of POCD, and evaluate the efficacy ofpostoperative application of Si ni tang and donepezil in POCD patients.Methods:48health aged male SD rats screened with the experimental requirements，weighing550-650g，were randomly divided into three groups：．Sini tang group(A group),donepezil group(B group) and the control group(C group). All of them took thepreoperative training of the Morris water maze and open field test in six consecutive days.Anesthesia with isoflurane splenectomy in the same condition The anesthesia process for4h. Postoperative stay awake rats before given medicines. postoperative After payattention to keep warm. Gavage for seven days. The1st day after operation conduct watermaze and open field test. On the one day before surgery, immediately after surgery,postoperative24h venous blood. After3,7,21d water maze and open field tests. Finallyfrom the brain and slicing, expression was detected by immunohistochemistry of β-AP.Determination of the content of blood ACTH, ALD, TNF-α, S-100β and NSE.Results: rats during the process of operation, stable vital signs, water mazeexperiment results show that, after operation first,3days, the rats of group a escape latencywas significantly shorter in group B and group C (P<0.01). Results: after1D, open fieldacross the lattice count, group A stand up times, defecation frequency and the other twogroups have obvious differences, both have statistical significance. For the TNF-α, ACTH,ALD, NSE, S100β index, three groups were increased significantly, C group than in A, Bgroup increased more significantly; deposition of β-AP and so.Conclusion: the results after the application of Sini tang can effectively inhibit theoccurrence of stress reaction after operation, reduce the deposition of β-AP in the brain;reduce the postoperative incidence of cognitive impairment; It can increase the expressionof hippocampus of rat choline acetyltransferase and nicotinic acetylcholine receptor application of donepezil after operation, also can reduce theβ-AP deposition in brain, and itmay play some role in prevention and treatment on postoperative cognitive dysfunction.
