[Chapters1] To establish kidney ischemia-reperfusion injury model in mice[Objective] Kidney ischemia-reperfusion injury in mice model is establishedsuccessfully, provide support for follow-up study[Methods]30male BALB/C mice were divided into2groups: the model group(n=24),control group(n=6). the model group: the left kidney ischemia was45min,right kidneywas removed,48h reperfusion, postoperative observed survival rate, measured serumcreatinine levels, kidney specimens for HE staining. control group:only opened,bilateral renal pedicle were separated, right kidney was removed, didn’t do any otherprocessing. postoperative measured serum creatinine levels, kidney specimens for HEstaining.[Results] The postoperative survival rate of model group was91.67%, serumcreatinine level was significantly increased, pathological injury of kidney was typical.[Conclusion] Kidney ischemia-reperfusion injury in mice model was establishedsuccessfully, postoperative survival rate was higher, renal function was significantlyimpaired,pathological injury of kidney was typical. conform to the requirements of theexperimental.[Chapters2] The study of effects of Salvianic acid A on kidney ischemia-reperfusioninjury in mice.[Objective] Clear the role of Salvianic acid A on kidney ischemia-reperfusion injury inmice[Methods]20male BALB/C mice were divided into5groups: the sham group(n=4),IRI group(n=4), IRI-15mg group(n=4), IRI-30mg group(n=4), IRI-60mg group(n=4).IRI group,IRI-15mg group,IRI-30mg group,IRI-60mg group: according to the operationway in Chapters1ischemia-reperfusion injury model was established. IRI-15mg group,IRI-30mg group,IRI-60mg group were intervened with Salvianic acid A, serumand kidney specimens were collected in postoperation, kidney tissue specimens weremade into paraffin section for HE staining, to observe the kidney pathological damagemicroscopically; measuring serum creatinine,blood urea nitrogen, to assess kidneyfunction of mice.[Results] The Scre and BUN levels of IRI-30mg group(113.95+2.91、48.44+1.81)、IRI-60mg group(86.12+3.13、40.63+3.49)were lower than IRI group(169.54+5.15、71.48+4.92), the results have statistical significance(P<0.001、P<0.001;P<0.001、P<0.001);compare the IRI-15mg group, the IRI-30mg group and the IRI-60mggroup, the difference have statistical significance (P<0.001、P<0.01;P<0.001、P<0.01);IRI-15mg group、IRI-30mg group、IRI-60mg group: pathological damagedegree in turn reduce.[Conclusion] Salvianic acid A can relieve kidney ischemia-reperfusion injury inmice.[Chapters3] The study of mechanism of action of Salvianic acid A on kidneyischemia-reperfusion injury in mice.[Objective] Prove the mechanism of Salvianic acid A that relieves kidneyischemia-reperfusion injury in mice[Methods] To measure the expression of IL-1β、IL-6、TNF-α with RT-PCR; to measurethe activity of myeloperoxidase with chemical colorimetry; to measure the proteinconcentration of nephridial tissue with coomassie brilliant blue method; to measure theactivity of superoxide dismutase with the xanthine oxidase method; to measure thecontent of malondialdehyde with glucosinolates barbituric acid method.[Results] The activity of myeloperoxidase(17.39+0.58)of IRI group,the expression ofIL-1β(2.35+0.06)、IL-6(2.38+0.06)、TNF-α(2.53+0.041) of IRI group were higherthan sham group(11.44+0.54、0.97+0.08、1.03+0.05、0.99+0.06),the results havestatistical significance(P<0.05、P<0.001、P<0.001、P<0.001);the activity of superoxidedismutase of IRI group(120.57+2.94)was lower than sham group(156.08+9.06), the content of malondialdehyde of IRI group(17.29+0.65)was higher than sham group(11.11+0.71), the difference has statistical significance(P<0.05、P<0.001、P<0.001、P<0.001). The activity of myeloperoxidase of IRI-30mg group(14.98+0.69), IRI-60mggroup(13.53+0.94)were lower than IRI group(17.39+0.58), the results have statisticalsignificance(P<0.05、P<0.05),to compare the activity of myeloperoxidase of IRI-15mggroup(16.44+0.63)and IRI group(17.39+0.58), the results have no statisticalsignificance(P>0.05);The activity of myeloperoxidase: IRI-60mg group <IRI-30mggroup<IRI-15mg group, the differences have statistical significance(P<0.05); theactivity of myeloperoxidase of IRI-60mg group was higher than sham group(11.44+0.54),the result has statistical significance(P<0.05); The expression of IL-1β、IL-6、TNF-α of IRI-60mg group(1.46+0.05、1.45+0.06、1.50+0.05)、IRI-30mg group(1.80+0.07、1.82+0.08、1.79+0.05)、IRI-15mg group(2.09+0.13、2.25+0.06、2.19+0.14)were lower than IRI group(2.35+0.06、2.38+0.06、2.53+0.041), thedifferences have statistical significanc(eP<0.05); The expression of IL-1β、IL-6、TNF-α:IRI-60mg group <IRI-30mg group<IRI-15mg group, the differences have statisticalsignificance(P<0.05); The expression of IL-1β、IL-6、TNF-α of IRI-60mg group werehigher than sham group(0.97+0.08、1.03+0.05、0.99+0.06), the results have statisticalsignificance(P<0.001、P<0.001、P<0.001); The activity of superoxide dismutase ofIRI-30mg group(131.08+3.96), IRI-60mg group(137.38+2.85)were higher than IRIgroup(120.57+2.94), the results have statistical significance(P<0.05、P<0.05),tocompare the activity of superoxide dismutase of IRI-15mg group(124.42+2.76)and IRIgroup(120.57+2.94), the result has no statistical significance(P>0.05);the contentof malondialdehyde of IRI-30mg group(15.43+0.58), IRI-60mg group(13.93+0.78)were lower than IRI group(17.29+0.65), the results have statistical significance(P<0.05、P<0.05),to compare the content of malondialdehyde of IRI-15mg group(16.58+0.65)and IRI group(17.29+0.65), the result has no statistical significance(P>0.05);The activity of superoxide dismutase: IRI-60mg group>IRI-30mggroup>IRI-15mg group, the differences have statistical significance(P<0.05、 P<0.05、P<0.05); The content of malondialdehyde: IRI-60mg group <IRI-30mg group<IRI-15mg group, the differences have statistical significance(P<0.05、 P<0.05、P<0.05); The activity of superoxide dismutase of IRI-60mg group was lower than shamgroup(156.08+9.06),the result has statistical significance(P<0.05); The content ofmalondialdehyde of IRI-60mg group was higher than sham group(11.11+0.71),theresult has statistical significance(P<0.05).[Conclusion] Salvianic acid A relieve kidney ischemia-reperfusion injury in mice byanti-inflammatory, antioxidant.
