Objective: To explore the changes and significance of the activation of IL-17/Stat3signaling pathway insepsis strengthen blood glucose by detecting the expression level of IL-17, IL-6and Stat3in the liver,lung tissue of sepsis rat of insulin target blood glucose management.Method:(1)We collected liver and lung tissues in the rat model of sepsis of different glucoseconcentrations prepared by modified CLP.Divided them into five groups: Sham operation group(Groupsham, n=7); Sepsisgroup(group CLP, n=7); Blood glucose control groupA (targetglycemic controlin4.4~6.1mmol/L, n=7); Blood glucose control group B (target glycemic control in group of6.1~8.3mmol/L, n=8); Blood glucose control group C (target glycemic control in8.3~10.0mmol/L, n=8). Scorethe liver and lung injury by histopathological observation. The expression of IL-17/Stat3(IL-17, IL-6andStat3) signaling pathway protein in the lung and liver tissue of septic rats in different glucoseconcentrations in different groups was detected by Non-biotin PV-6001two-step, to observe its expressiondifference and explore the role and significance of IL-17/Stat3signal pathway protein in different glucoseconcentrations in septic liver, lung injury in rats. Statistical analysis was performed using SPSS17.0statistical software for statistical analysis, using χ2test, group were compared using the Fisher exact test,Spearman rank correlation test. Results:(1) Different blood glucose concentration and CLP lung injury insepsis rats were significantly heavier than liver injury group, CLP in the liver, lung injury is the mostserious, glycemic control, A, B and C of liver, lung injury change with elevated bloodglucose.Concentration injury was gradually aggravated: A group <B group <C group. The lung tissue wasmainly manifested as pulmonary septal thickening,alveolar capillary congestion, alveolar wall,inflammatory cell infiltration and interstitial pulmonary edema. Liver damages were liver cell hydropicdegeneration, fatty degeneration, height liver sinus congestion and interstitial inflammation.(2) Lung tissuepathological score showed: in CLP group and glucose compared to the control group and sham group, thedifferences were statistically significant (P <0.05), the degree of lung injury was gradually aggravated.Hepatic tissue pathological score showed: in CLP group and blood glucose control group compared withthe control group had significant difference (P <0.01); especially the two indexes of hydropic and fattydegeneration of liver cells. In general, the degree of lung injury was significantly greater than that of liverinjury.(3) In group CLP expression rate were the highest expression of IL-17, IL-6, p-Stat3in liver and lung tissues, compared with the sham group, there was significant difference in C group and CLP groupblood sugar control difference(P<0.05), suggesting that IL-17, IL-6, p-Stat3control of C group and CLPgroup showed high expression in severe injury of blood glucose blood sugar control; A group or B groupand sham group showed no significant difference, suggesting that intensive glycemic control can reduce theliver, lung injury;(4) IL-17expression and IL-6and Stat3showed a positive correlation between IL-17,IL-6,and had Stat3and liver, lung injury score significant correlation, suggesting IL-17/stat3signalingpathway may play an important role in the occurrence of sepsis, development, and its expression level ishigher, the degree of liver, lung injury of important organs more. The activation of p-Stat3is related to thedamage of liver and lung tissue. And the injury more serious, the expression gets higher.Conclusion:(1)When the blood glucose concentration of sepsis increases, the damage degree of liver andlung is aggravated,and especially lung tissue injury is significant.(2)The expressions of IL-17, IL-6andp-Stat3are related to lung and liver injury in sepsis, suggesting that IL-17/Stat3signaling pathway mayplay an important role in the development of sepsis;(3)The protective effects on the liver and lung injury inrats with sepsis is the most obvious when the blood glucose is controlled in4.4~6.1mmol/L comparedwith6.1~8.3mmol/L and8.3~10.0mmol/L.
