| With the improvement of people’s living standard and change of lifestyle, the incidence of diabetes mellitus(DM) increased year by year and has become the third severe disease after cardiovascular disease and cancer which would seriously harm human health. On Nov.14,2011, the5th World Diabetes Day, the statistical results promulgated by China showed that more than94.2million adults in our country have diabetes(more than90%of them are type2diabetes),which make our country the largest population of diabetics in the world. What’s more is that the diabetics become younger, the prevalence of diabetes between25and34increased eight-fold compared with1994. The prevalence of diabetes of men and women over20were as high as10.6%and8.8%,and the prevalence of pre-diabetes is as high as15.5%. The diabetes reserve force which means pre-diabetes populations is as much as148million.In the face of such a long-course and easy cause complications disease, in the face of more than100million patients in the early stage of type2diabetes, we should control it with early prevention and early treatment. And this is fit for the thought of prevention before disease onset and preventing disease from exacerbating in preventive treatment of disease. Therefore we made the preventing treatment of disease as our guiding thought and do our research with pre-diabetic animal model.ObjectiveTo explore a rat model of early stage of type2diabetes, and do the research of clinical effective Chinese herbal compound and its disassembled prescription based on this model, remedy the rats with Wenyang compound, Wenyang double-drug compatibility, Bushenyang compound, Bushenyang double-drug compatibility, negative compound(compound without thermal Chinese medicine) and melformin as the positive control medicine. We observe the drug effects in the weight of rats once a week, the Lee’s index, the obesity level, the index of blood glucose the index of bloot lipid and body temperature, and then we do research in the expression of GLUT2of liver tissue and GLUT4of muscle tissue of the rats. To discuss the therapeutic effect of the Chinese herbal compound on early stage of type2diabetes rat model, establishing a theoretical basis in drug development and clinical practice.MethodsMale SPF Sprague-Dawley rats, weighed80-120g, were randomly chosen10as normal control group, which were fed with normal standard diet, the rest were fed with high fat and sugar diet. After12weeks, selected the rats which are20%overweight, detect the FBG, TCH, TG, FINS of10heaviers and10lighters of the overweight rats to verify the rat model of early stage of type2diabetes success. Then they were divided into7groups which are model group, melformin group, negative compound group, Bushenyang double-drug compatibility group, Bushenyang compound group, Wenyang double-drug compatibility group and Wenyang compound group. All rats were treated for8weeks. Their body weights were recorded once a week. Their FBG and body temperature were be examined at the end of4th week. The OGTT experiment would be took at the end of6th week. At the end of8th week, the rats were fasted for12hours, their FBG and body temperature were be examined. The serum TG, TC, HDL-C and LDL-C levels were measured by automatic biochemical analyzer, FFA were measured by ELIASA, GHbAlc were measured by ELISA Reader, insulin were determined by radioimmunoassay. After animals’execution, liver, kidney and adipose tissue were collected and weighted, then the visceral fat mass and its index were calculated. GLUT2and GLUT4protein levels were measured by western blotting.ResuIts1. The exploring of establishing a rat model of early stage of type2diabetesFeeding rat with high fat and sugar diet for12weeks could simulate greasy diet and simulate clinical symptoms of early stage of type2diabetes. This model could obviously increase TCH, TG, INS of rats and increase FBG a little bit, this is quite close to the diagnosis of early stage of type2diabetics. Thus this rat model of early stage of type2diabetes has definite feasibility.2. The effects of glucose and lipid metabolism of early stage of type2diabetes rat modelWenyang compound could reduce body weight, body fat, FBG, GHbAlc, hepatic glycogen, INS, TCH, TC, LDL-C, FFA in the meanwhile increase HDL-C as well as or better than melformin. Those compounds with Qingre an Huashi medicine in it are also effective in reducing bloot lipid, but not good at reducing bloot glucose. While Bushenyang medicine isn’t effect in neither bloot glucose nor bloot lipid.3. The effects on GLUT2signaling pathways in liver tissue and GLUT4signaling pathways in muscular tissueWenyang compound could up-regulate GLUT2and GLUT4protein levels and its effect is superior to melformin. The effects of other group are not obvious.ConclusionThis study successfully established a rat model of early stage of type2diabetes. The results show that Wenyang compound could effectively reduce weight of model rat, correct the disorder of blood glucose and blood lipid, improve insulin resistance, up-regulate GLUT2and GLUT4protein levels while Bushenyang compound did a under-performance. The results could feedback and guide the clinical medication. |
