ObjectiveObservation of effect of Cordyceps polysaccharide combined MSCs after transplantation and extracellular matrix degradation. Preliminary study of Cordyceps polysaccharide synergistic mechanism of MSCs transplantation in rats with liver cirrhosis. Discussion of TCM and MELD score,MELD-Na score between the relations,"syndrome" objective,"syndrome" to provide an objective basis for judgement of severity and prognosis.Methods12weeks of continuous subcutaneous injection of carbon tetrachloride induced liver cirrhosis in rats, after injecting,35rats were randomly divided into5groups, each group of7.MSCs transplantation group and MSCs transplantation+Cordyceps polysaccharide group separately from thetail vein injection of GFP labeled MSCs2×106, Cordyceps polysaccharidegroup and MSCs transplantation+Cordyceps polysaccharide group at seconddays after transplantation given Cordyceps polysaccharide gastric perfusion. All rats were sacrificed after transplantation, serum was collected for liver function, hepatic fibrosis indexes detection, determination of liver tissue hydroxyproline content, and HE staining and Masson staining, immunohistochemical detection of MMP-13and TIMP-1.2008July to2011July were collected medical record data, analysis in the194case patients at admission to the MELD score and MELD-Na score and syndrome type of traditional Chinese medicine.Results(1) The model control group, MSCs transplantation group, Cordyceps polysaccharide rats serum ALT activity was significantly higher than that in the normal group(KO.05);MSCs transplantation+Cordyceps polysaccharide group ALT level slightly higher than normal group(P>0.05);and the model control group rats compared to the treatment group, ALT activity was lower than that of model group(P<0.01);MSCs transpl antation+Cordyceps polysaccharide group ALT activity was lower than that of MSCs transplantation group and Cordyceps polysaccharide group(P<0.05).(2)Rank test statistic analysis of serum AST activity, normal group,model control group, MSCs transplantation group,Cordyceps polysaccharide group, MSCs transplantation+Cordyceps polysaccharide group Mean Rank were18.64U/L,19.29U/L,19.57U/L,16.86U/L,15.64U/L, AST in every activity to compare difference not to havestatistical significance(P>0.05).(3)MSCs transplantation group, Cordyceps polysaccharide group,MSCs transplantation+Cordyceps polysaccharide rats serum ALB levels were lowerthan the normal group(P<0.05), significantly higher than that in the modelgroup(P<0.01); the treatment groups there wasno statistically significant difference between the content of ALB(P>0.05).(4)Compared with the normal group, model control group and the treatment group, LN. PCIII. HA levels were significantly higher(P<0.01), as compared with model group, the treatment group in the three index levels were lower (P<0.01), MSCs transpl antat ion+Cordyceps polysaccharide group were transplanted with MSCs group and Cordyceps polysaccharide group in comparison, there were statistically significant (P<0.05).(5)MSCs transplantation group,.MSCs transplantation+Cordyceps polysaccharide group of hydroxyproline content was higher than normal rats(P<0.01), but was significantly lower than that of model group (P<0.05);MSCs transplantation+Cordyceps polysaccharide group than that in MSCs transplantation group, hydroxyproline content decreased and has statistical significance(P<0.05).(6)Compared with model group, the treatment group,MMP-13expression was significantly increased(P<0.01), TIMP1(P<0.05),decreased expression of transplanted MSCs+Cordyceps polysaccharide group and MSCs transplantation group and Cordyceps polysaccharide group compared with statistically significant difference (P<0.05).The six group of syndromes of MELD, MELD-Na score mean average water as follows:the yin deficiency of liver and kidney-yang deficiency of spleen and kidney,blood stasis>wet-hot>water wet resisiance>liver qi stagnation, and statistical significance (P<0.05) Conelusion1Cordyceps polysaccharide with protection, repair damaged liver cells, improving liver function, inhibit collagen synthesis, promote the degradat on of ECM with liver cirrhosis, treatment effect.2MSCs transplantation can treat liver cirrhosis, to a certain extent improve liver function, promote collagen degradation, reduced collagen deposition, with remission or reversal of hepatic fibrosis.3Cordyceps polysaccharide and MSCs transplantation in treatment of liver cirrhosis with synergistic effect, the two union application, can the advantage is complementary,synergistic, improve liver function, promote collagen degradation, reduced collagen deposition, alleviate or reverse liver fibrosis.4liver qi stagnation, moisture resistance, damp-heat stagnation, blood stasis, spleen kidney yang deficiency, yin deficiency of liver and kidney in six groups of syndromes in patients with posthepatitic cirrhosis reflects the development stage, various types and severity of disease and prognosis related to cirrhosis of the liver, spleen kidney yang deficienc, yin deficiency of liver and kidney in two syndromes of liver cirrhosis with serious illness, poor prognosis.
