Polymer nano-preparation of antitumor drug is an important research directionof new chemotherapeutic drug. Chemotherapy is one of three major cancer treatment,it has irreplaceable position in the field of cancer therapy. However, many cancerpatients have to stop chemotherapy because of intolerable chemotherapy toxicity ofheart, vomiting, nausea, hair loss and other side effects in clinical, which greatlyinfluence the treatment and quality of life of the patients. A important reason forserious side effects of chemotherapy drug is the drug toxicity and no selectivity, itkills tumor cells as well as strongly damage the normal cell and tissue function in thesame time. Through developing targeted agents of antitumor drugs, improving themetabolism kinetics and biodistribution of drugs, so as to get the drug concentratingin the tumor cells and to reduce the side effect of antitumor drugs dignificantly.Polymer nano carrier can enhance the targeting ability of antitumor drugs andanti tumor effect, one of the important reasons of which is nano carrier improve themetabolism and biodistribution of antitumor drugs. There is a great relationshipbetween the physical and chemical properties of nano preparation and metabolism andbiodistribution of nanoparticles.Although regarding to factors of affecting metabolism and biodistribution ofnanoparticles become more and more, according to the study of liposomes have beenmore careful, with a lot of valuable results. However, for the fate of the polymer nanopreparation in animal, there are still a lot of questions to be answered. Such as:howthe nanoparticles metabolic in vivo, how the physical and chemical properties affectthe metabolism and biodistribution of nanoparticles,and metabolism and biologicaldistribution of nanoparticles in different tumor types. The answers to these questionsare meaningful on the study of novel tumor targeting carrier of drugs. This paper explored how the physical and chemical properties affect the metabolism andbiodistribution of nanoparticles in cellular and animal level, through the change of thegrafting amount of the targeting groups or polyethylene glycol which can prolong thecirculation time of nanoparticles in vivo respectively.As the results proved,we havesuccessfully synthesized two types of tumor probe, RGD-PLG-CS andPEG-PLG-IR830, which suit for the photoacoustic imaging.we also found that withthe increase of RGD and PEG,the accumulation of poly(glutamic) acid nano carrierwere increased in tumor.
