Object:Gastrointestinal related tumors serious threat to human health.Existing tumor treatment results are still unsatisfactory. Mainly due to theearly detection of gastric cancer is difficult, and the existing drugtreatment is not ideal. So it is urgent to find a new and effective targetedtumor imaging and drug carriers to improve the prognosis ofgastrointestinal tumors. The purpose of this experiment is to use phagedisplay techniques, screening the proteins produced by gastric cancertissue, to find gastric cancer specific antigens which can provide newmolecule carriers to detect tumor and tumor targeted therapy.Case selection: Randomly selects5patients in our department whoreceived the radical gastrectomy (pathologically confirmed gastricadenocarcinoma). We get gastric carcinoma and normal gastric mucosaabout3grams each during the surgery, frozen.Method: Picking out Gastric cancer tissues and normal gastricmucosa, after mincing, cells lysing and other procedures, we get proteinorganizes. Picking out an appropriate amount of each protein organizesafter two-dimensional electrophoresis, silver stained, we get2-DE gelsrespectively.We give2-DE gels an electric shock to turn to the PVDF membrane.We select phage by the gastric cancer tissue organizes, and multiplicate them after infection E. coli. Linking the Phage antibody prepared in theprevious experiments with gastric cancer tissue and normal gastric tissue,and detected by Western blot. We find the different protein spotscompared with two groups of PVDF membranes and select one differentprotein spots out of silver stain plastic to analysis by Mass spectrometer.Result: Through comparative analysis, we found that seven antigens ingastric tissue which can’t be found in normal gastric mucosa. This studynot only provides candidate biomarkers for the early diagnosis of gastriccancer, but also provides a new direction for the treatment of gastriccancer.
