Purpose:This study was conducted to determine the neuroprotective effects of activeLigustrazine hydrochloride on SH-SY5Y cells injured by H2O2and explore theunderlying molecular mechanisms.Methods:1. Establishment of the model of oxidative damage in SH-SY5Y cells induced byH2O2;2. Using MTT assay to determine cell proliferation;3. Using FCM to detect induced-apoptosis;4. Based on Western blot method to test the expression of MMP-9protein;Conclusions:1. The optimal concentration for H2O2-induced damage in SH-SY5Y cells modelis200μM, and the optimal time is four hours. The best protection time forLigustrazine hydrochloride is two hours.2. The Ligustrazine hydrochloride could be used to protect H2O2-induced damagein SH-SY5Y cells, and the effect of the protection is positive correlation withthe dose used.3. This experiment revealed that the protective effects of ligustrazinehydrochloride against H2O2-induced damage may through the down-regulationof MMP-9protein. |
