Objective:In this study, Liquid chromatography-mass spectrometry was used to describethe metabolites of blood samples and urine samples from patients with gastric cancerand healthy controls. Then find the different components between the samples andfind the biomarkers.Methods:1. Sample preparation: The blood and urine samples were processed bycentrifuging for minutes, the upper phase was used for further analysis.2. LC-MS analysis: The samples were analyzed by HPLC-TOFMS. Then weget the metabolic patterns of samples of blood and urine and related data. Then findthe differences between them.3. Data analysis: Several statisticalanalysis such as principle componentanalysis, t-test, hierarchical cluster analysis and partial least squares-discriminantanalysis were used to analysis the data between the patients and healthy controls.Then found significant differences between the two metabolites, and verification ofthese metabolites in gastric cancer patients and healthy people discriminating ability.Results:1. In the study of blood samples, the results show that in the PCA score plot,gastric cancer patients and healthy people can be well distinguished. Then use the ttest, clustering analysis, PLS-DA to screen different metabolites. Then identify thestructure of then in the HMDB,21species in total was identified positive andnegative mode. Finally, after graphing the ROC curve,14important metabolites wereselected with high sensitive.2. We get21species in blood and27species in urine which show thedifferences between the patients and healthy control. In from which,14in the bloodand3in the urine were important which show high sensitive and specificity. Theresults of this study demonstrated that these metabolites could distinguish the gastriccancer patients and healthy people and could be used as potential biomarkers. Wefound Quinone and Acetyl-N-formyl-5-methoxykynure-namine in both blood andurine,so these two metabolites could be specific metabolites of gastric cancer.
