Objective:The effect and pharmacodynamics of’Chuan Qi Fang’ compound formulation compared to single compound ophthalmic solution on the retinal degeneration models, provided an insight on new type of ophthalmic solution and the choices on dosage level to the treatment of retinal degeneration.lt also served as a foundation and basis for other Chinese medicine extract ophthalmic solution formulation in other ophthalmic diseases.Methods:1. Using water extraction and alcohol precipitation method to prepare the’Chuan Qi Fang’ as an extract and after which dilute the extract with distilled water in the required proportion. The PH value of the solution containing active compound of Astragoloside and Tetramethylpyrazine in the ratio of1:1were also adjusted. Experimentation of using SD rats given eye stimulation, the’Chuan Qi Fang’ ophthalmic solution was administered20ug,3times a day,7days consecutively. The indicators are the overall health condition, retinal irritation symptoms, examination of the rats histopathology, which the condition and symptoms are recorded daily.2. The retinal degeneration rats model is induced using sodium iodide. The assessment of the treatment of’Chuan Qi Fang’and single compound positive control were observed though electroretinograph and histopathology of the eyes.The SD rats models were injected with40mg of sodium iodide, randomized into the’Chuan Qi Fang’group and single compound positive control group and given treatment. The electroretinalgraph, histopathology examination, RPE cell spread preparation and pathological examination and HE staining were carried out on day7,14to observe the retinal function and histopathological changes. TUNEL method was also used to examine the apoptosis of the different layers of retina cells.3. Using Immunohistochemistry to observe RPE cell and the change in the expression of the photoreceptor cell protein, and hence discuss the pharmacodynamics of the’Chuan Qi Fang’. After modelling, the eyeballs of the rats were taken out from the day7or day14. After conventional frozen sections, changes in the RPE65and the expression of opsin were observed using Immunohistochemistry. Results:1. Sodium Iodide can lead to retinal degeneration which included RPE cells and extent was relative to the dosage and time.2. Sodium Iodide had a greater impact on Rod-ERG, Cone-ERG and Flick ERG in relation to the retinal function. Therefore Sodium Iodide’s impact of inducing apoptosis on RPE and cones were selective.3. deducing from the result of RPE cell spread preparation, TUNEL and RPE65protein expression, Sodium Iodide could lead to the decrease of RPE65protein expression, increase in the cell apoptosis and induce apoptosis in RPE cells, which was also dosage dependant.4.’Qi Chuan formula’ophthalmic solution could reduce the extent of the damage on retina.Conclusion:1.’Chuan Qi Fang’ophthalmic solution had treatment effect on sodium iodide induce retinal degeneration model rats in which Astragoloside had the better effect in anit-apoptosis and protecting retinal tissues and followed by Tetramethylpyrazine.2.’Chuan Qi Fang’ophthalmic solution and Tetramethylpyrazine ophthalmic solution both had positive effect in the treatment while Escuilin and Digitalisglycosides ophthalmic solution had no effect.
