Objective:Studying the probable repair mechanism of electroacupunture combined with aerobic exerciser intervent the interacation of PICK land the GluR2subunit of AMPAR,which may improve cognitive function in rats of chronic ischemic congnitive impairment,by observing the effects of electrocupuncture baihui,shenshu,dazhui,Xuan zhong combined with aerobic exerciser on the rats’congnitive function,the expression of PICK1and GluR2in the hippocampus,the hippocampal tissue Hestaining.Which provide us with more reliable erperiment for the clinical application of cerebral ischemia.Methods:Sixty male SD rats were randomly divided into group A(blank group), group B(sharm operation group), group C(modle group),group D(aeribic exercise group),group E(electroacupuncture group) and group F (electroacupuncture combined with aerobic exercise group).Rats of chronic cerebral ischemia model was made by permament bilateral common carotid artery ligation except group A and group B. group B only separeted the carotid artery ligation. After8weeks of electroacupuncture baihui,dazhui, shenshu,yongquan,or combined with swing train used Morris water maze to test the congnitive function in rats.Histolagical changes in the hippocampus were observed by light miscrosope,the content of PICKl and GluR2were detected by immunohistochemistry.Result:1.Water maze test shows that rats’escape latent period prolonged but space exploration period shorten after molding which explain the model of chronic cerebral ischemia is successful.After8weeks treatments,the groups of electroaccupunture combined with aerobic exercise, escape period shorten but space exploration period prolongde.the differences were significant between them(P<0.05).2.To observe hippocampus neurons under light microscope after HE staning:the shape of hippocampus neurons were normal in the blank group.Hippocampus neuron were not complete in the model group:cells are spare and disorder,nuclears are pyknosis.the damage in the groups of D、E、F were neat,the structure of neurons were complete relatively.3. Electroaccupunture combined with aerobic exercise can reduce the expression of PICK1in rats of chronic cerebral ischemia. The differences were significant between the the groups of D、E、F and the modle group(P<0.05).4. Electroaccupunture combined with aerobic exercise can increased the expression of GluR2rats of chronic cerebral ischemia. The differences were significant between the the groups of D、E、F and the modle group(P<0.05).Conclusion:1. Electroaccupunture combined with aerobic exercise can improve the cognitive function in rats of chronic cerebral ischemia,which are benifical for cerebral function.2. Electroaccupunture combined with aerobic exercise can reduce the expression of PICKl in rats of chronic cerebral ischemia, but increase the expression of GluR2,which may effect the interaction on them and benifical for cognitive function of cerebral.
