Objective:By studying the effect of antler growth factor on vascular endothelial cells of rats withischemic limbs(VEGF)、microvessel density (MVD) and the influence of blood enzyme,explore of the effect of pilose antler on angiogenesis of rat Ischemic hind limb, lookingfor effective treatment for clinical treatment of hind limb ischemic diseases.Methods:108SD rats using randomized experiment method is divided into: normal group, modelgroup,high,middle,low dose pilose groups,compound salvia table grounp, deer antler,compound salvia tablet group and model group were established the left leg branch offemoral artery ligation and stripping out, due to hind limb vascular ischemia model, normalgroup exposed femoral triangle, only not ligation femoral artery and its branches. velvetantler group began in0.1ml/100g take pilose antler,1times a day, for14consecutivedays. Normal group and model group to fed with the distilled water.One day before surgery, postoperative temperature measured daily rat hind legs on bothsides, After3days,7days,14days, rats in each group were randomly selected six,conducted in peripheral blood creatine kinase (CK), lactate dehydrogenase (LDH),aspartate aminotransferase (AST) determination; take surgery lateral gastrocnemius, lowerlimb ischemia recovery observed capillary structure calculated capillary density (unitsvision Aizen area/unit field area); left leg hematoxylin-eosin (HE) staining pathology was observed between the number of blood vessels in skeletal muscle cells; determinationof VEGF protein expression in muscle tissue; observed after the first14days of each groupleft lower limb necrosis number of cases.Results:1.Three days postoperatively, ischemia rats hind limbs temperature is reduced, the colorwhite, not seen gangrene, wound scab over, did not see obvious hemorrhage,7days afterwound healing,14days after the scabs fall off, see model group1case of ischemic hindlegs black foot pad,1nail to fall off, the low-dose group antler see1case of ischemic hindleg foot mat in black.2.Rat serum enzyme assay results: by Kruskal-Wallis H test: normal group bloodenzyme values each time, there was no significant difference after3days, each AST,CK, LDH value more than group were higher than blank group, model group, AST,CK values more deer antler velvet antler low-dose group of dose and a significantrise in salvia miltiorrhiza group (P <0.01); After7days, each AST, CK, LDH valuesdrop, velvet antler high, medium and low dose group and salvia miltiorrhiza groupcompared with model group decreased significantly (P <0.05), among them, velvetantler is the most significant the high dose group (P <0.01). Velvet antler high,middle dose group was significantly lower than CK values model group, high dosegroup is lower than dose group;14days postoperatively, low dose and effect was CKvalues with the model group had significant difference (P <0.05), the AST, CK,LDH values showed a trend of slow decline earlier, no obvious difference with7days postoperatively, and3days after significant difference (P <0.05).3. Microvessel density (MVD) measurement results: by Kruskal-Wallis H test:7days after surgery, antler MVD in the high-dose group was significantly higher thanthe model group (P <0.05),14days after surgery, antler MVD in the high dosegroup was significantly higher than the model group and low dose group (P <0.05).4.VEGF expression assay result: VEGF expression by Kruskal-Wallis H test:after three days, antler high dose, antler high, middle dose, expression of salvia miltiorrhiza group compared with model group, significant difference (P <0.05),velvet antler highest high dose group of VEGF protein expression values;7daysafter each VEGF expression began to decline, velvet antler high, middle dose andsalvia miltiorrhiza group decreased significantly (P <0.01);14days postoperatively,velvet antler high dose group of VEGF expression increased significantly in the othergroup (P <0.05), velvet antler in high dose group, velvet antler dose group, danshengroup differences reduce postoperative3days expression significantly (P <0.05).5.Left leg hematoxylin-eosin (HE) staining pathology: the normal group ratmuscle cells arranged in neat rows, between the muscle fibers have a certain amountof blood vessels; muscle cells of rats in the model group atrophy, interstitialhyperplasia, an increase in the number of blood vessels compared to the controlgroup; antler high-dose group rat muscle cell contraction lighter, the number ofblood vessels between the muscle fibers compared with the model group and more;antler, and between the low-dose group of muscle fibers increase in the number ofblood vessels, but less than the high-dose group; Salvia piece group slightlyincreased vascularity.Conclusion:Through determination of each index of rat, antler can promote lower limbischemia of lower extremities ischemia model of rats VEGF expression, promoteangiogenesis, alleviate limb ischemia. Effect of high dose group is better than thelow dose group, it may be able to mobilize endothelial progenitor cells and velvetvalue-added differentiation and migration.
