Objective: The research is to investigate the effects of stellate ganglion block onisolated cardiac function of Langendorff isolated heart perfusion device in SD rats, andto supply the experimental information for scholars to improve the cardiac functionclinically and reduce cardiovascular events during the peri operation period.Methods:24male SD rats which aged8to10weeks old and weight180to220g wererandomly devided into three groups: left stellate ganglion block group (groupLSGB),right stellate ganglion block group (group RSGB) and and control group (groupC).The left and right chest muscle were seperated bluntly after the anestnesia of rats byintraperitioneal injection.The stellate ganglion was finded along theparaesophageal,then buried the pipe and sewthe skin.The stellate ganglion blockexperiment was conducted after seven days,then observe whether Horner’s syndeomewas occured and inject the drug continously for two weeks.The Langendorff isolatedheart perfusion device was applied to set up isolated heart perfusion model on SD ratsafter the successful establishment of stellate ganglion block model.Rats wereintraperitioneally injected with heparin,fixed in supine position,quickly open the chest and remove the heart.The heart was hang on the Langendorff isolated heart perfusiondevice after isolating the aortic in the4℃cold K-H liquid.The heart was perfused bythe K-H liquid,and then record related index of cardiac function after determining thecoronary flow at the end of the balance,such as heart rate(HR),left ventriculardeveloped pressure(LVDP),left ventricular end-diastolic pressure(LVEDP),maximumrates of increase and decrease in left ventricular pressure(±dt/dtmax).At the sametime,take the supernatant after centrifuging the heart by steps of addition ofstandard,sampling,adding antibodies,washing and adding substrate. Detectepinephrine(EPI), norepinephrine(NE), neuropeptide tyrosine(NPY),vasoactiveintestinal peptide(VIP) of the myocardial tissue by ELISA.Results:1. LVDP,+dt/dpmax,-dt/dpmax,CF and HR of group RSGB were increasedwhen compared with group C(P<0.05or P<0.01) while LVEDP decreasedstatistically significantly(P<0.01).LVDP,+dt/dpmax,-dt/dpmax and CF of group LSGBwere increased when compared with group C(P<0.05or P<0.01) while LVEDPdecreased(P<0.05).LVDP,+dt/dpmax and-dt/dpmax of group RSGBwere increasedwhen compared with group LSGB(P<0.05) while LVEDP decreased(P<0.05).2.EPI,NE,NPY and VIP of group RSGB and LSGB were increased when comparedwith group C(P<0.05or P<0.01).EPI,NE,NPY and VIP were increased whencompared with group LSGB(P<0.05).Conclusion:1.Stellate ganglion block can promote isolated cardiac function of SD ratsthrough enhancing myocardial systolic and diastolic function.2.Both sides of stellate ganglion block have different effects on heart function in SD rats as the sympathetic nerve innervation area of the right and left stellate ganglion isdifferent.And the right stellate ganglion block is better on enhancing the heart function.
