Yixin tongmai oral liquid is experience prescription used in our school affiliated hospitalas the treatment of pectoral stuffiness and pains, qi deficiency and phlegm stasis.Take Under the theories of Traditional Chinese Medicine and Pharmacy as the guidance,and with the help of the modern pharmaceutical technology,The pre-clinical pharmacy of Yixin tongmai suppository has been researched.According to the function,treatment and the characters of the effective component of the herbs,the preparation process is designed,Taking the extract rate of volatile oil as index, the extraction of volatile oil were selected with the single factor investigation method,water-extract were selected with the orthogonal test which use Calycosin-7-glucoside and dry paste rate to evaluate.single factor investigation method was used to optimize purification methods among water sinking method,physical centrifugation and alcohol sinking method by taking clarify degree of solution and content of Calycosin-7-glucoside as indexes.at the same tine,the condensation,purification and shaping condition were screened.At last the preparation of yinxin tongmai oral liquid were determined as follows:chuanxiong and xiebai were used to extracted300mL aromatic water by adding15times of water,the liquid was mixed with lOmL Polysorbate80and the mixture was filtered, the dregs were reserved to be extracted with huangqi and danshen in10times of water for3times,each time contained1hour;The filtrate was merged and the extraction solution was concentrated to relative density1.05～1.15under the temperature of40～50℃. The concentrate was centrifuged and concentrated to600mL. Finally1.5g sorbic acid and the aromatic water were added in the concentrate and the pH was adjusted to5.5～6.5, some water was added to1000mL.The quality standard of Yixin tongmai oral liquid was set up, which includes the thin layer chromatography identification of huangqi,danshen and chuanxiong,and the determination method of Calycosin-7-glucoside was assayed by HPLC,which contained the system methodology. According to the content of Calycosin-7-glucoside in different batches of samples,the content limit was established:per mL of yixin tongmai oral liquid contained Calycosin-7-glucoside shall not be less than0.014mg.According to the quality standard,the preliminary stability research had kept by by accelerated tests, the results indicated that yixin tongmai oral liquid was steady and the wrap was suitable.All of the studies above indicate that the preparation procedure of yixin tongmai oral liquid is reasonable, the quality standard is controllable, the sample is steady on the whole as well.
