Objective:Based on the current phenomenon of obese children’s increasing, we estabish an animal model of diet-induced obesity pregnant rat and their offspring. By the model we can simulate diet-inducedobesity pregnant mothers and the effect on their next generation’s obesity and artery atherosclerosis lesions. And we want to explore pathophysiological mechanisms of the effect.Methods:We establish the animal model of diet-induced obesity pregnant rat. Pregnant rats were randomly divided into four groups:normal diet group (bassic feed100%), the high-energy diet during pregnancy (basic feed80%,10%lard,1.5%cholesterol,0.5%bile salts,8%sucrose). Early pregnancy (10days early in pregnancy) high-energy diet group, late pregnancy (10days late in pregnancy) high-energy diet group. To explore the relationship between diet-induced obesity pregnant rat,offspring’s atherosclerotic lesions, blood insulin and aquaporin-7.Results:1. The body weight, body fat, and Lee’s index of the high-energy diet during pregnancy group and early pregnancy high-energy diet group are more significant increase than the datas of normal diet group when the offspring rats are4weeks and9weeks old.2. The level of insulin secretion and insulin sensitivity index of the high-energy diet during pregnancy group and early pregnancy high-energy diet group are significant increase than the datas of normal diet group when the offspring rats are4weeks old (P<0.01). There are no significant increase of the level of blood glucose and lipids between these groups. All of these items are significant increase than the items of normal diet group when the offspring rats are9weeks old.3. By immunohistochemical method, we can confirme that the expression of aquaporin-7are significant reduction in the high-energy diet during pregnancy group and early pregnancy high-energy diet group than in the normal diet group.4. The area of atherosclerotic lesions in abdominal aorta are significant Increase in the high-energy diet during pregnancy group, early pregnancy high-energy diet group and late pregnancy (10days late in pregnancy) high-energy diet group than in the normal diet group.5. By real-time PCR, we can confirme that the expression of aquaporin-7are significant reduction in the high-energy diet during pregnancy group and early pregnancy high-energydiet group than in the normal diet group. Conclusion:1. The incidence of offspring’s obesity and atherosclerotic lesions are significant increase in the high-energy diet during pregnancy group and early pregnancy high-energy diet group.2. By the expression of aquaporin-7, the pregnant rat can adjust the fat metabolism and the fat content of artery of offspring. And the incidence of cardiovascular disease of offspring is increase.
