Codonopsis lanceolata is widely distributed in East Asia and is used in traditional medicines area. Modern pharmacological experiments show that, C. lanceolata has various significant abilities such as anti-tumor, anti-aging and antioxidant effect. This paper is aiming to extract and isolate the monomer compounds from the EtoAc extraction part and n-Butanol extraction part from C. lanceolata, screening for which one has the anti-tumor activities from those compounds and set the foundation for purity determination for the further research.Root samples were extracted with80%EtoAc and60%EtoAc respectively and then were extracted with EtoAc and n-Butanol. The water solution was passed through D101column, silica gel column and Sephadex LH-20column to give five fractions. The structures of those compounds were identified on the basis of physical-chemical constants and spectral data. Compound2was screened by MTT methods to elucidate the inhibitory effects on the growth of human liver cancer cell line HepG-2.1. C. lanceolata was chromatographed on silica gel column and Sephadex LH-20column repeatedly to give five compound, which were determined based on spectral data such as’H-NMR and13C-NMR and identified by comparing the spectral data with their authentic data. They are:dodecan-2-one(Ⅰ)3-0-β-D-glucuronopyranosyl-3β,16a-dihydroxyolean-12-en-28-oic acid28-0-a-L-Rhamnopyranosyl-(1'2)-[β-D-glucuronopyranosyl-(1'3)]-a-L-arabinopyranosyl ester, named lancemaside D(Ⅱ)、Syringin(Ⅱ)、3-0-β-D-glucuronopyranosyl-3β,16α-dihydroxyolean-12-en-28-oic acid28-0-β-D-xylopyranosyl-(1'3)-β-D-xylopyranosyl-(1'4)-α-L-rhamnopyranosyl-(1'2)-[β-D-glucopyra-nosyl-(1'3)]-α-L-arabinopyranosyl ester, named lancemaside B(Ⅳ)、compound5has not identified yet.2. The MTT experiment show that, the inhibitory ratio is much higher than control group when the concentration during40~100μg/mL, and the higher the concentration of lancemaside D the better effect of the inhibition. |
