| Objective:To study the quality standard of Ocimum basilcum. Methods:according to the"Chinese Pharmacopoeia"2010version of the appendix part,to inspect moisture, total ashand volatile oil of Ocimum basilcum; optimization thin layer identification condition oflinoleic acid in Ocimum basilcum and determining the β-sitosterol content of10batchesOcimum basilcum by gas chromatography;Using gas chromatography-mass spectrometryanalysis of volatile oil components in the Ocimum basilcum; By single factor experimentand orthogonal experiment, optimized the extraction process of total flavonoids fromOcimum basilcum; Using rutin as the standard, determining the content of total flavonoidsin Ocimum basilcum by UV spectrophotometric method of10batches Ocimum basilcum.Results:This experiment determined initially with methanol, chloroform (7:3) as solvent,ultrasonic extraction and petroleum ether, ethyl acetate (37:3) for a system,1%vanillinsulfur acid as the chromogenic agent of linoleic acid thin layer identification, andpetroleum ether as solvent, ultrasonic extraction and petroleum ether, ethyl acetate (5:1)for a system,10%of phosphomolybdic acid ethanol solution as the chromogenic agent β-sitosterol thin layer identification, spots are clearly visible. Using gas chromatography of10batches of samples for central Asia oleic acid determination, it is concluded that allareas of Ocimum basilcum the linoleic acid content were between0.27~0.29mg/g. Usinghigh performance liquid chromatography (HPLC) method for10batches of samples for β-sitosterol determination,64compounds were identified from the Ocimum basilcumessential oil, consistent with the literature. it is concluded that all areas of basil herbs its β-sitosterol content were between0.20~0.32mg/g. Preliminary determine the bestextraction technology of total flavonoids in Ocimum basilcum; And rutin as a standard, uvspectrophotometry of10batches of Ocimum basilcum samples to determine the content offlavonoids, all parts is concluded Ocimum basilcum of the flavonoids content between0.0450~0.0785mg/g.Conclusion: The TLC method of linoleic acid and β-sitosteroldetermined the preliminary experiment is feasible, with good reproducibility, can be usedas a method of TLC identification of Ocimum basilcum. This study compared the contentdetermination of total flavonoids in the samples of10batches of Ocimum basilcum,linoleic acid and β-sitosterol, and preliminarily determines the optimum extraction process of total flavonoids in Ocimum basilcum; provide reference data for the futurestudy of Ocimum basilcum quality criteria establishment and effective parts. |
