Tumors are a kind of serious disease. Each year the number of people losing their lives because of tumor is only second to the number of people dying of cardiovascular disease. It has become the leading of death to human life after cardiovascular disease, and the high morbidity and mortality of lung cancer have already drawn many scientists’ attention. In recent10years, the morbidity and mortality of lewis lung carcinoma have always ranked number one among the malignant tumors in many areas of our country. Though the research on tumor is extensive, the research report in the relationship between the mechanical properties of cells and lewis lung carcinoma is few. We explore the mechanism of cell in process of its pathological changing through the observation and research on the variation between the lewis lung carcinoma and normal lung cells.Cell traction force(CTF) is a very important physical parameter of cells and has a close relationship among inflammation, wound healing, angiogenesis, signal transduction and so on. Based upon the research methods and basis of the mechanics of lewis lung carcinoma at home and abroad, this article research the mechanical characters of lung cancer before and after the changing.Object:Through the contrast of the traction to the cells, explore the changing rules of cancer cell in the mice before and after the cancer variation and the influence of cancer variation of cell on the cellular mechanics properties.Methods and conclusions:1. Culture cells.Obtaining the target cell, put it in the CO2incubator and culture it there. After three or four days, change the medium and remove the adherent cells immediately. Observe the development form of cell with invert microscope, chose the better one for expanding culturing. Multiply the next generation every three days and put it aside for experiment by the time most of the cells growing in good condition.2.The sum of cyclic peptide dyeing. Through the sum of cyclic peptide dyeing technology, take out the cells growing well and dye the normal lung cells and the lewis lung carcinoma.The result shows that the microfilament cells are not in order but in distorted shape after cancer variation and present a energy contained state.3.The SMA protein detection. Using fluorescent antibody techniques to dye the lung cells and lung tumor cells within mice respectively, finding that the SMA protein is declining and in scattered distribution after the lung cells’tumor variation in the mice.4.The measurement of the cell traction force. Using CTFM (Cell Traction Force Microscopy), namely, microscope tracking method. Respectively detect the cellular traction before and after the cells’cancer variation in the mice and find that the cellular traction reduces by49%after the changing. The average amount reduce from552pa to281pa.The average amount of cells’projection superficial area reduce950um2to691um2,reducing by27%.The biggest displacement of the cell on the matrix reduces from2.5um to2.2um.The result of this research reveals the form, internal conformation, and the change of the protein expression and traction. It provides the preliminary basis for the deep research on the mechanism in the process of lung cells’ the cancer variation and it means a lot.
