Objective: Observed Yishenhuoxue Ruanjian Turbidness recipe on renal interstitialfibrosis in rats fibroblast transforming growth factor protein (TGF-βlmRNA), affect theexpression of hepatocyte growth factor protein (HGFmRNA), as renal interstitial fibrosisprovide the basis for prevention.Methods: Surgical ligation of the unilateral (left) ureteral rats (UUO rats) as a modelof renal interstitial fibrosis.The108male SD rats were randomly divided into four groups,sham operation group, model group, benazepril group, oral group, each27.The treatmentgroup received Yishenhuoxue Ruanjian Turbidness prescriptions (Astragalus AngelicaRadix Salvia Chuanxiong gun a bead peach rhubarb charcoal Plantago amomum licorice)formulated to contain crude drug concentration20.6g/ml decoction treatment given to eachgavage2ml; benazepril group2ml/kg/d gavage, model group and sham group pouredsaline.Treatment underwent enucleation left kidney removed kidney tissue, pathologicalchanges were observed in the light of kidney tissue; application expressionimmunohistochemical detection of renal tissue of rats of TGF-β1and HGF;usingmolecular biology techniques to check real-timePCR detection of renal tissue TGF-βlmRNA, HGFmRNA of expression.Results:(1)Rat body weight the sham operation group increased significantly, ingood spirit; rat model group, weight loss, listlessness, decreased activity; symptomscompared with the model group rats orally relatively light.(2)Renal pathological changesnaked eye view: rat kidney sham group reddish brown, normal renal tissue; model groupsignificantly increased compared with normal renal volume, surface thinning of corticaland edema; oral benazepril group and the control group than in the model group relief.Microscope Appearance: the sham operation group structure of normal kidney tissue, noinflammatory cell infiltration, no focal fibrosis, no tubular dilatation; rat model of renaltubular dilation group, renal interstitial visible brown color change, with large number ofinflammatory cell infiltration, focal fibrosis obvious; between oral renal tissue interstitialfibrosis, inflammatory cell infiltrationsmall tube dilation had improved andreduced.(3)Compared with the normal control group,serum BUN,Scr,CRP and Alb werehigher in model group,but there was no obvious difference between the normal control group and the model group(P﹥0.05);(4)Serum BUN,Scr,CRP and Alb levle, there was noobvious difference between the medicine methods group and the benazepri group(P﹥0.05);(5)Compared with the model group, the oral group, benazepril group two in withrenal of rats TGF-β1were significantly lower (P <0.01), the expression of HGFsignificantly increased (P <0.01).(6)Compared with the model group, oral group,benazepril kidney tissue of rats in the control group mRNA and protein expression of TGF-β1were significantly increased (P <0.05), HGFmRNA and protein expression wassignificantly decreased (P <0.05).Conclusion:(1) Yishenhuoxue Ruanjian Turbidness UUO model can reduce thetubular atrophy, inflammatory cell infiltration, and the degree of expansion of thetubules.(2) Yishenhuoxue Ruanjian Turbidness may directly or indirectly inhibit theoverexpression of TGF-β1, and increase the expression of HGF in renal tissue, preventionand treatment of renal interstitial fibrosis, so as to achieve the purpose of protecting renalfunction.
