purposeThe recent literature on the treatment of lung cancer using hyperthermiacombined with chemotherapy or radiotherapy more, but the three combinedtheoretical study reported less. The experiment will hyperthermia, chemotherapyand radiotherapy combined effects of the three lung adenocarcinoma A549cells,the survival rate was observed to provide an experimental basis for the clinicaltreatment of lung cancer three combined.methodsShow a logarithmic growth phase and in good condition randomized lungadenocarcinoma A549cells,①Control group: After replacing the culturemedium for48h in an incubator;②Chemoradiotherapy group: containing thedrug concentration is1.25×10-10mol/L of docetaxel cultured24h, then give6GY6MVX high energy irradiation, cultured24h;③Hyperthermia combinedwith radiotherapy and chemotherapy group: containing the drug concentration of1.25×10-10mol/L of docetaxel cultured24h, giving different temperaturehyperthermia, including38℃,39℃,40℃,41℃,42℃,43℃, at a constanttemperature water bath according to experimental treatment groups after1hdesired temperature heating, and then give6GY6MVX high energy irradiation,cultured23h. These three groups according to the specified time after thetraining is completed, measured by MTT viability of A549cells, and thendetected by flow cytometry apoptosis of A549cells, and Western Blotapplications detect the expression of Bcl-2and Bax. Results(1)By MTT assay of A549lung adenocarcinoma cell survival: Whenhyperthermia temperature≤39℃,the survival rate slightly higher than thechemotherapy group compared with the chemotherapy group, the difference wasnot statistically significant (P>0.05); After as the temperature increases, thatis, when the temperature≥40℃,lung adenocarcinoma A549cell viabilitydecreased, compared with the chemotherapy group, the difference wasstatistically significant (P <0.05). When hyperthermia temperature≤39℃when the survival rate is higher than the A549lung adenocarcinomachemotherapy group, because the temperature has not been reported to reducethe role of radiotherapy and hyperthermia have been reported related to thetemperature may be reduced when paclitaxel≤39℃effect on lungadenocarcinoma A549cells[30], it is considered that the temperature may reducethe role of docetaxel for lung adenocarcinoma A549cells, pending furtherstudy.(2)Measured by flow cytometry apoptosis of A549display: put apoptosiscompared with chemotherapy significantly after43℃combined radiotherapyand chemotherapy.(3)Applications Western Blot detect the expression of Bcl-2and BaxShow: put after chemotherapy was significantly reduced compared with theexpression of Bcl-2after43℃combined radiotherapy and chemotherapy, whichsignificantly increased the expression of Bax.Conclusions(1)When hyperthermia temperature≤39℃higher than the survival rateof lung adenocarcinoma A549chemoradiotherapy group, suggesting thattemperature may reduce the effects of docetaxel in lung adenocarcinoma A549.(2)When the temperature≥40℃hyperthermia when combined withchemotherapy, the treatment effect is good. When temperature is43℃, thetreatment effect is the best.
