With the continuous improvement of living standards, and diabetes are rising year byyear. According to the survey show that the people of type2diabetes are about more thanthe total number of95%. Diabetic nephropathy (diabetic nephropathy, DN) is one of themost common and serious chronic microvascular complications of DM. The mainpathological changes of diabetic nephropathy is kidney high perfusion, high filtration,glomerular basement membrane thickening and mesangial extracellular matrix areaaccumulation, leads to diffuse and nodular glomerular sclerosis, and appears clinicalsymptoms of hypertension, proteinuria and renal dysfunction. Traditional view is thatchronic complications of diabetes are a relative or absolute lack of insulin-inducedhyperglycemia. Recent studies have found that Diabetes were closely related tohemodynamic changes, glucose metabolism, gene polymorphisms, ECM degradation, lipidperoxidation, cellular and vasoactive factors, inflammation, genetics, growth factors andother factors.tTG is a class of Ca2+-dependent enzyme, can produce a variety of reactions throughan acyl transfer effect in the body, and promote the crosslinking of collagen and otherECM ingredients, form a solid structure, and thus resist to degradation.UTI is out of isolated and purified glycoprotein fresh urine from healthy adult males.It is composed of143amino acids, and its relative molecular mass is67KD. It belongs tothe protease inhibitor. It has the inhibition to trypsin, α-chymotrypsin, serine proteases andgranulocyte elastase, hyaluronidase, thiol enzyme, plasmin, and other enzyme. In addition,it has a stable lysosomal membrane, can remove oxygen free radicals and inhibit therelease of inflammatory mediators. Ulinastatin was mainly used in treatment of acutepancreatitis, chronic recurrent pancreatitis in clinical.In this experiment, Wistar rats were randomly divided into six groups, including thenormal control group, STZ-induced model group (25mg/kg), STZ+Captopril group(captopril,5mg/kg/d), STZ+Ukrainian (high, medium and low,25mg/kg/d,20mg/kg/d,15mg/kg/d). The change of rat kidney was measured by HE staining. The collagen contentof kidney tissue was detected by Masson trichrome. Expressions of tTG were detected by immunohistochemistry and real-time PCR.The results showed that fasting blood sugar of rat is stable and maintained in a stablerange (4.9to6.1). Blood glucose levels in model group was significantly higher than innormal group (P <0.05). The glucose in Captopril group and Ukrainian group decreased ascompared with the model group (P <0.05). The results of HE staining, Masson staining andexpression of tTG showed that ulinastatin may reduce the expression of tTG, and thus playan important role in prevention and treatment of diabetic nephropathy.
