Objective: To establish a model of osteoblast cultured in vitro and to observe andinvestigate the effect of Astragaloside A on osteoblastic proliferation and differentiation.Preliminary discuss the possible cytological mechanism of Astragaloside A for repair ofbone defects. Methods:This periment used cell culture technology to culture the calvariumosteoblasts. Osteoblastic differentiation was examined by reserved phase-contrastmicroscope, scanning electron microscope, alkaline phosphatase staining and Alizarin redstaining. Cells of successive transfer culture were seeded and cultured in24well cellculture cluster. After confluenced, the extracts of Astragaloside A was applied to cellculture, then the ALP content in the culture fluid was detected. Cells were seeded andcultured in96well cell culture cluster. After cultured by the extracts of Astragaloside A,the function of osteoblastic proliferation was detected by MTT method. Results: theextracts of Astragaloside A with different concentration(1ug/ml、10ug/ml、100ug/ml) canincrease the ability of osteoblastic proliferation; the extracts of Astragaloside A withconcentration of10ug/ml and100ug/ml can increase the ability of osteoblasticdifferentiation.Conclusion: the extracts of Astragaloside A with certain concentration canimprove the osteoblastic proliferation and differentiation. Perhaps the effect ofAstragaloside A is a kind of cytological mechanisms of the Chinese medicine functions ofpromoting the concrescence of bone fracture and preventing osteoporosis.
