ObjectiveVitamin D can not only regulate calcium and phosphorus metabolism, but also closely relate to the occurrence and development of cardio-cerebrovascular disease. So far, some studies have found that NF-кB can be activated when ischemic stroke happened. After activated, the NF-кB can participate in the inflammation, and exacerbate the damage of the brain. If we can prove that the pathway of the NF-κB has been inhibited or blocked up by vitamin D, we can have the conclusion that vitamin D is a protective factor to ischemic stroke. Therefore, in this study, we detect the activity of the NF-кB to prove our conjecture.Methods72 male SD rats were randomly divided into sham-operative group, control group, VD-deficient group and VD-sufficient group. Rats in sham-operative and control groups received normal diet. Rats in VD-deficient group received VD-deficient diet. Rats in VD-sufficient group received VD-sufficient diet. The MCAO model was made after each group pretreated for 10 weeks. Surgical procedures of sham operation group was the same as the above, but no thread inserted. Neuroethology evaluations were made for each group of rats, and the cerebral pathological conformation were observed by HE staining. The expressions of NF-кB detected by immunohistochemical and Western blotting.Results1、The effect of VD on neurological severity score: Compared with the control group, we found that the symptoms of neurological deficit in VD-deficient group are more aggravate, and the.score is much higher(P<0.01). While, the symptoms in VD-sufficient group are more alleviated than control group, and the score is much lower.2、The effect of VD on HE staining of brain tissue after focal cerebral ischemia::Compared with the control group, we found that the damage degree of neuron in hippocampus CA1 area in VD-deficient group is much more aggravate(P<0.01). While, the damage dgree in VD-sufficient group is alleviated than control group.3、The effect of VD on the expression of NF-κB P65: Compared with the control group, the VD-sufficient group can inhibit the expression of NF-κB P65 by immunohistochemical and Western blot methods. While, the expression of VD-deficient group is more than the compare group(P<0.01).Conclusions1、VD can remarkblely alleviate the symptoms of neurological deficit, and improve the damage degree of neuron pattern of focal cerebral ischemia rats, which play a protective role to the rats.2、Deficent of vitamin D can increase the activity of NF-κB, and may cause the cascade of inflammatory, which can exacerbate the damages of the brain.3、VD can inhibit the acitivition of NF-κB, and suppress the inflammatory response. The protective role of vitamin D is played by inhibiting the NF-κB path way.
