Object: The regulation of local immune system and its mechanism were discussed by studying the effect of sympathetic nerve on the signal transduction pathway mediated by MCP-1.Method:(1) Sympathetic blocking experiments: 60 healthy female mice were divided into control(A)and experimental(B) groups.Each subgroup had 5 mice. For A group, 0.2ml 6-hydroxydopamine(100mg/kg) was injected into their enterocelia at 9 am everyday for 5 days. While at the same time, the same doses of physiological saline were injected into the B group. The mice uteruses were extracted separately on day 1, 3, 5, 7, 9 and 14 at 3 pm.(2) NE injection experiments after sympathetic blocking: 20 healthy female mice were divided into 4 groups.All the mice were injected with 6-OHDA to block the sympathetic nerve.And then for experimental group, two injections were performed with NE at every morning and evening at the doses of 0.02μg, 0.2μg and 2μg. While for control group, the same doses of physiological saline were injected. All the mice uterus were extracted on the 5th day at 3pm. Detect the expression of CCR2 in mice uterus tissue through immunohistochemistry.And use in situ hybridization to detect the mRNA expression of CCR2 and MCP-1.ELISA was used to detect the content of MCP-1and IP3 in mice uteruses.Results:(1) Compared with the control group, none of the indicators had significant change on the 1th day after the injection of 6-OHDA. Since the 3th day,all the indicators were raised. And the trend of change is increased first and then decreased. In each part of the uterine tissue,endometrium expresed more MCP-1 and CCR2 and the changes in endometrium were most obvious after the injection of 6-OHDA and NE.(2) Compared with the control group,after the injection of NE, the expression of MCP-1 and CCR2 was significantly enhanced in low concentration group.It also showed a significant high expression in medium group,but it’s lower than low concentration group.High concentration of NE had no significant effect on the expression of MCP-1 protein, but it can significantly enhence the expression of MCP-1mRNA.And also it had no significant effect on the expression of CCR2, the expression of CCR2 in endomrtium even showed a significantly down regulation compared with the control group.(3)Compared with the control group,The concentration of IP3 was raised after the injection of 6-OHDA. And it reached a significant difference on the 5th day. After the injection of NE, the expression of IP3 was raised. And it reached extremely significant level in low concentration group while achieve significant level in medium concentyation group.Conclusion:(1) The activation and migration of immune cells(macrophages) in the uterine tissues were regulated by the sympathetic nerve, and this effect may be achieved by the signal transduction pathway mediated by MCP-1.(2) The regulation of the sympathetic nerve to the immune cells in the uterus is related to the secretion of the transmitter NE, and the function of NE maybe related to its concentration, and there maybe a bidirectional regulation mechanism.(3) MCP-1 and its recepter CCR2 can achieve the chemotaxis of immune cell through IP3 signaling pathway.
