Phthalate esters, commonly referred to as PAEs is predominantly used as a plasticizer which main effect is to soften plastic and enhance the flexibility of plastic, it’s toxic effect has gained our attention. In all PAEs, dibutyl phthalate, commonly referred to as DBP, is one of the most serious pollution, exposure to the highest level in body. DBP will be hydrolyzed to Mono-n-butyl phthalate which is also called MBP. PAEs urine metabolites test shows that DBP metabolites has the highest content of MBP, DBP metabolites studies have shown that it has a variety of toxicity, can affect the male animal reproduction and development, but its mechanism is not very clear, still needs further research to clarify.This paper explores the reproductive toxicity of MBP, toxic effects and the mechanism of injury from the individual level, organization level and molecular level. Through test weight and viscera coefficient, testicular tissue pathology slice observation and sperm density and sperm deformity rate of detection on mice after MBP exposure, To evaluate testicular tissues extracted damage from exposed MBP. Through doing the detection on the change form testicular tissue oxidative stress indicators(ROS,MDA,GSH) and energy metabolism related enzyme activity (LDH, SDH), testicular tissue growth and sperm works related gene (Sox9, DAZL, drmtl) expression,to explore the possible mechanisms of MBP produce toxicity. In normal condition and under the condition of using protectant to protect testicular tissue, through the research on mice testis tissue damage and toxicity after MBP exposure, to explore the possible mechanisms of testicular tissue injury in mice from MBP, And by adding melatonin as a protective agent, to study the melatonin in MBP after exposure on protection effect and protection mechanism of testicular tissue.The experimental result shows that. With the exposure of the MBP, the concentration group of 100 mg/kg/day can lead to testicular tissue injury, born seminiferous tubule atrophy, tube cavity various living sperm cells decreased significantly, and the concentration group of 200 mg/kg/day will lead to the further testicular pathological changes in mice, arrangement of interstitial cell sparse, dividing unusually, a substantial reduction in the number, sperm density and sperm deformity rate also appeared a significant decline. At the concentration group of 100 mg/kg/day and 200 mg/kg/day, ROS level and MDA content in testicular tissue have a significant rise,while GSH level, LDH and SDH activity have a significant decline. Begin from 50 mg/kg/day, the expression of related genes Sox9 and DAZL has a significant decline. When add the melatonin (50 mg/kg/day) as a protectant, and MBP at the concentration group of 200 mg/kg/day, melatonin can relieve mice testicular tissue damage, reduce the rate of sperm deformity in testicular tissue, sperm density restoration, also can reduce oxidative stress, improve energy metabolism related enzyme activity, so that testicular tissue damage from the MBP will get a certain degree of improvement.
