| Experiment of Ethylnitrosourea(ENU)-induced mutagenesis in mouse was carried out in this research. At last 3 kinds of ocular mutant strains were acquired according to the genetic testing, clinical diagnosis and pathological observation; Then complished preliminary localization of the mutant gene in one case of mice with small eyes by the method of microsatellite marker linkage analysis. The work is summarized as follows:1.120 of C57BL/6J(B6) adult male(8-10 weeks) were treated by ENU injection, then mated these mice(generation 0,G0) with female normal mice. 2782 generation 1(G1) mice were bred, and 65 mice with ocular abnormal phenotypes were filtrated.According to the genetic testing, clinical diagnosis and pathological observation,3 genetic cases were reserved and the mice showing respectively corneal opacity, small eyes and iris abnormality.2. Focusing on small eyes mice as the study material, we mapped the mutant gene in the method of linkage analysis and found the LOD was 2.53 between the D8Mit4 and the gene, so linkage was confirmed between them.Above of all, we bred 3cases of ocular abnormal mice that showing similar phenotype with that in some human diseases, providing good materials for research of new drugs; the mutant gene in small eyes mice was preliminary located in D8 chromosome, supplying firm foundation for fine location. |
