The Infection And Genotyping Of Toxoplasma Gondii In Leukemia Or Lymphoma Patients In Guizhou Province

Objective: To explore the molecular-epidemiology of Toxoplasma Gondii infection in hematological cancer patients in Guizhou province. Serological test and Multiple Nested Polymerase Chain Reaction-Restriction Fragment Length Polymorphism(Mn-PCR-RFLP) was applied in this study. Method: 1. Peripheral blood were collected from 150 leukemia patients, 150 lymphoma patientss and 110 healthy people. 2. The specific Ig G, Ig M and CAg were detected by enzyme-linked immuno sorbent assay(ELISA).3. The 529 base pairs DNA fragment of Toxoplasma Gondi were amplified by PCR. 4. Genotying of Toxoplasma Gondi was confirmed by Mn-PCR-RFLP using ten genetic markers including c22-8, c29-2, L358, PK1, SAG2(5`-SAG2, 3`-SAG2, alt. SAG2), BTUB, GRA6, SAG3, Apico and SAG1. Result: 1. Positive rates of Toxoplasma Gondi infection were 8.18%(9/110), 18.67%(28/150)and 21.33%(32/150)in healthy people, leukaemia and lymphoma patients, separately. The Positive rates of Ig G were 6.36%(7/110), 16.00%(24/150) and 20.00%(30/150), Ig M were 0.91%(1/110), 2.67%(4/150)and 1.33%(2/150), CAg were 0.91%(1/110), 2.00%(3/150) and 0.67%(1/150) in three groups. 2. Genotyping of Toxoplasma Gondi was abort because the 529 base pairs DNA fragment of Toxoplasma Gondi was negative in all samples. 3. The Mn-PCR-RFLP results confirmed that the genotype of Toxoplasma Gondi strains abtained from animals in Guizhou province belong to Toxo DB#9, named as “ Chinese 1 ”. Conclusion: 1. The positive rates of Toxoplasma Gondi infection in hematological cancer ptients was higher than healty people. Toxoplasma Gondi infection is common in hematologic tumors patiens. 2. It was confirmed that the genotype of Toxoplasma Gondii isolated from animals in Guizhou province is “ Chinese 1 ”.
