Objective:By tests in animal model and clinical observation, the experiment is designed to study the regulation of Xiangshaliujunzi Decoction on immunosuppression induced by diarrhea spleen deficiency and intestinal micro-ecology dysbiosis, reveal the mechanisms of preparation in treatment of diarrhea spleen deficiency animal model and children, and provide a basis for clinical application of Xiangshaliujunzi Decoction to diarrhea spleen deficiency patients.Methods:1. Animal model.It is using large dose of decoction of rhubarb retention enema lavage on mice to do the experimental diarrhea spleen deficiency animal model-making and drug intervention which can observe the general situation of experimental mice, detect spleen weight index of mice, detect the change of erythrocyte immune C3 b receptor rosette and IC rosette, detect bifidobacterium, lactobacillus, bacteroides, enterococcus, enterobacter of stool specimens, and analyze the influence of decoction of rhubarb retention model-making and the effect of Xiangshaliujunzi Decoction on the immune function of mice and intestinal flora.2. Clinical observation. children with diarrhea spleen deficiency were divided into positive control group and Xiangshaliujunzi Decoction group for clinical control treatment and observation. Ig G, Ig A, Ig M, CD3, CD4 and the salivary amylase, the urine D-xylose excretion rate and the changes of stool SIg A were detected before and after treatment in the children, which is to validate the treatment effect of Xiangshaliujunzi Decoction on the children with diarrhea spleen deficiency.Results:1.After model-making, the indicators of lactobacillus, bacteroides, enterobacter, enterococcus and bifidobacterium in the intestinal tract of intestinal flora in diarrhea spleen deficiency mice model show a significant difference(P<0.05). Splenic index and the index of erythrocyte C3 b receptor rosette of mice model were lower than those in normal group. After the treatment with Xiangshaliujunzi Decoction, the index of bifidobacterium, lactobacillus and bacteroides were significantly increased, the difference was statistically significant(P <0.05). But the index of enterococcus and enterobacter were significantly reduced, and the difference was statistically significant(P <0.05). Before and after treatment, the index of bifidobacterium and bacteroides in the mice stool specimens of high dose group have a remarkable difference in Statistic meaning(P <0.01). Splenic index and the index of erythrocyte C3 b receptor rosette were significantly improved, and the difference was statistically significant(P <0.05) and IC rosette rate had no obvious difference in Statistic meaning(P >0.05).2. The total effect rate of Xiangshaliujunzi Decoction group is 100% in clinical control experiments, and the difference of clinic effect compared with positive control group was statistically significant(P <0.05). There were statistically significant difference of Ig G in the two groups before and after treatment(P <0.05). There were no statistically significant difference of Ig A and Ig M before and after treatment(P <0.05). CD3, CD4 and CD8 were lower than those in normal group(P <0.05,P <0.01), which improved after treatment, particularly for CD3, CD4(P <0.05). Compared treatment group of Xiangshaliujunzi Decoction to positive control group, the salivary amylase, the urine D-xylose excretion rate and stool SIg A of two groups were significantly improved after the intervention treatment, and the difference was statistically significant(P <0.05).Conclusion:Xiangshaliujunzi Decoction which is prescription of replenishing vital energy used in the experiments improve intestinal micro-ecological environment and establish the intestinal natural biological barrier protection by advancing immune indexes of diarrhea spleen deficiency animal model and children, which play a therapeutic effect of diarrhea spleen deficiency.
