[Objective]More and more attention is paid on the role which stroma plays in tumor carcinogenesis now. Based on previous research results, this study aimed to explore the effects of over-expression of Periostin, a stroma protein, on Nasopharyngeal carcinoma and its mechanism. [Materials and Methods] Lentiviral expression vector of POSTN and GFP were transfected into 6-10 B cells. Western blot and PCR were used to confirm the transfection effects. Then the effects of over-expression of Periostin on nasopharyngeal carcinoma were examed by MTT, migration, invasion and gelatin zymography. At the same time the expression of Periostin and Integrin-αvβ5 were detected by IHC in NPC and NNM, and the expression of Periostin and Integrin αvβ5 was also detected in 6-10B-POSTN cells, 6-10B-GFP cells and 6-10 B cells. The expression levels of Periostin and Integrin αvβ5 in primary NPC were ananlysed with Spearman statistics. [Result] With the stable transfection 6-10 B cells(6-10B-POSTN), their biological characteristic analysis was performed as follows:1)The MTT assay showed that up-regulated Periostin in 6-10 B cells was associated with a marked increase of cell growth compared with the contorl groups,; 2) Scrach test and Transwell chamber invasion assay showed that up-regulation of Periostin expression in 6-10 B cells was associated with increased in vitro cell migration and invasion; 3) MMPs activation tested by gelatin zymogram suggested an increase in MMP-2 and MMP-9 activity in cultivated supernatant of 6-10B-POSTN compared with 6-10B-GFP and 6-10 B. Periostin mainly expressed in the stroma, and the expession level of Periostin obviously stronger in the stroma of nasopharyngeal carcinoma than normal nasopharyngeal mucosa. Integrin mainly expressed on the membrane, the expression levels of Integrin-αvβ5 in primary NPC and 6-10B-POSTN cells were significantly higher than those in NNM and 6-10B-GFP, 6-10 B cells. The expression levels of Integrin-αvβ5 in NPC showed positively correlated with the expression of Periostin(r=0.682, p=0.000). [Conclusion]Results from above experiments suggegsted that Periostin plays an important role in regulation of cell proliferation, the activity of MMPs, cell migration and invasion probably by combining with Integrin-αvβ5. Our results will be helpful to provide a new idea for finding stromal targets of tumor therapy.
