| Objective:Observe rat tibia fracture healing process in the callus tissue of rats I collagen type and Vascular endothelial growth factor (Vascular endothelial growth factor, VEGF) expression, in-depth research and analysis of bFGF and the FS complex on fracture healing promoting,and to provide some experience for the clinical treatment of the fracture.Methods:Selecting 45 healthy male SD rats 5 months only, randomly divided into bFGF+FS, FS, NS three groups, each group of 15, only three groups of rats were separately cross shape in the upper left tibial fracture, rat tibia fracture model is established. bFGF+FS group of animals every day on the left side of the gastrocnemius muscle note bFGF+Fibrin gel (Fibrin sealant, FS) compounds, to partial put to death; Group FS animals every day on the left side of the gastrocnemius muscle bone peptide injection of 1 ml, and partial to be put to death; Group NS animals every day on the left side of the gastrocnemius muscle physiological saline injection of 1 ml, to batch to be put to death. Animal limb after three group were 2 weeks with a special small splint fixation, respectively in the building after 2 weeks,4 weeks,6 weeks 3 partial execution cycle experimental animal specimens, each 5 random kill rats in each group, only line X-ray observation of fracture healing, histological staining were observed under light of osteoblasts in bone scabs, immunohistochemical staining line I collagen type and VEGF immunohistochemical detection.Results:1. X ray film results:The same point in time after fracture, X-ray shows bFGF+FS group of callus generation most, the most number of osteoblast, collagen closely packed, fracture healing was significantly better than that of group FS and group NS.2. histological observation:Hematoxylin and eosin (HE) staining were observed under light, according to bFGF+FS number of bone cells slightly more than FS group, significantly more than group NS.3. immunohistochemical results:Collagen type bFGF+FS, FS group of â… dyeing image grey value is low, and goes on with the experiment cycle numerical bFGF+FS downward trend, show â… high expression of collagen type; Group NS grey value is high, which indicates that â… expression of collagen type is relatively low. Same time between different groups of collagen type â… grey value difference was statistically significant (P<0.05), group bFGF+FS and group FS, there are significant differences between group bFGF+FS and group NS (P<0.05), there are significant differences between group FS and NS group (P<0.05). The average optical density of VEGF group bFGF+FS image value (OD value) is high, is on the rise over time,6 weeks peak; group FS VEGF OD value of the image is slightly lower in group bFGF+FS, is on the rise over time, VEGF group NS image OD value is low, not obvious rise over time, OD value higher than that of groupFS, NS, group bFGF+FS had significant difference (P<0.05), between FS and NS groups have significant difference OD value, was statistically significant (P<0.05).Conclusion:1. BFGF in the course of fracture healing of rats can increase content of collagen type â… , inducing VEGF formation, stimulates the fracture healing of rats.2. The effect of bFGF fracture repair of rat superior to bone peptide. |
