Objective To detect the sensitivity and specificity of PNL2 monoclonal antibody and factors that affecting the dyeing rate of PNL2 in melanoma, to explore the meaning of the auxiliary diagnosis of melanoma. Methouds Using immunohistochemical SP method to detect 47 cases of melanoma (11 cases of mucosa,24 cases of acromegaly,12 cases of non-acra) and 10 cases of normal skin paraffin embedding in the organization,Then according to the expression of PNL2,S100 and HMB45 cases,analyzing their relationship with the clinical pathological characteristics. Results Of PNL2’s staining positive rate was 87.23%(41/47) in melanoma group,10%(1/10) in normal skin, respectively.There is the difference between these groups, with statistically significant (P<0.05). The sensitivity and specificity of PNL2 were 87.23%(41/47) and 90%(9/10). There are Age and grade of CLARK of PNL2 for melanoma with staining’s positive influence, the difference was statistically significant (P<0.05). It’s nothing to do with these foctors of Gender, Nationality and the metastasis of lymph node (P>0.05). Conclusion There is universal high specificity about PNL2 for melanoma and can be a good choice of auxiliary diagnosis of melanoma, especially it is more suitable for dyeing auxiliary diagnosis than of other immune markers that having a low positive rate in mucosal melanoma.
