Preparation And Application Of Reference Extracts From Forsythia Suspense

Forsythiae Fructus, the fruit of Forsythia suspensa (Oleaceae) is a climbing plant. Forsythiae Fructus is one of the most famous Chinese herbal medicines listed in the Pharmacopoeia of the People’s Republic of China. Which is widely distributed in Shanxi, Henan, Shandong, hubei, Sichuan, Jiangxi provinces of China, with heat clearing and detoxicating, detumescence effect. Chemical composition including caffeoyl glycosides, lignans, pentacyclic triterpene and volatile oil, etc., in which caffeoyl glycosides and lignans are the main bioactive components responsible for the various biological activities of Forsythiae Fructus. This topic mainly aims at the preparation and application of the extracts from Forsythiae Fructus, the experiment is divided into three parts.1. Preparation of reference extract:The first part of article was ultrasonic extraction forsythia with methanol and then on macroporous resin, silica gel and ODS column chromatography separation, the five kinds of chemical composition, identified were Lianqiaoxinside A, Forsythiaside A, (+)-Pinoresinol-β-D-glucoside, Phillyrin and Phillygenin.2. Application of reference extract:The second part of this article the HPLC method is adopted with octadecyl silane bonded silica as filler,10% acetonitrile-90% water as mobile phase, gradient elution, flow rate of 1.0 mL/min, detection wavelength of 280 nm and 330 nm, in 65 minutes, get five good separating degree of chromatographic peak. Five chromatographic peak of Lianqiaoxinside A, Forsythiaside A, (+)-Pinoresinol-β-D-glucoside, Phillyrin and Phillygenin.Forsythia suspensa reference extract study is presented in this article. Using Forsythia suspensa reference extract instead of standard determination of the contents of Lianqiaoxinside A, forsythoside A, pinoresinol-β-D-glucoside, phillyrin and phillygmin from 41 batch Forsythiae Fructus,32 batch Forsythia suspensa extract and 10 batch Shuanghuanglian oral liquid samples. And proved the feasibility of the method is simple, has a good reproducibility and accuracy. After statistics analysis meets the requirements, so make sure Forsythia suspensa reference extract instead of standard is aviable. Implements the quality control of the element of Forsythiae Fructus indicators, and overall quality control of traditional Chinese medicine (TCM) and the implementation of standards is of great significance.3. Determine the content of four components in Forsythiae Fructus by QAMS:selection of phillyrin as reference substance because phillyrin is one of the main active ingredients of Forsythiae Fructus, and the reference substance stability comes easily. Relative correction factor in the evaluation of different chromatographic column, liquid chromatograph, the influence of different laboratories, RSD is less than 5%, that of the correction factor between different types of compound has good credibility; Using QAMS and ESTD for determination of samples from different sources, QAMS and ESTD have no significant difference between the content of the results(P>0.05), showed that QAMS can be realized under the condition of the products is the lack of a variety of component content determination.
