| Objective:To observe the influence of oligodendrocyte precursor cells transplantation on motor function and medullated fibers morphology of experimental allergic encephalomyelitis rats.Methods:I cultured rat’s brain cortex in vitro, using separation and purification method for the separation of oligodendrocyte precursor cells with oscillation, appraisal cells by immunohisto-chemistry. I selected forty healthy rats for experiment randomly and divided them into normal group, model group, normal saline group, treatment group. I made clinical score for the model group, normal saline group, treatment group rats. About one week later, the treatment group rats were given ntravenous injection of Oligodendrocyte Precursor Cells. Then I observed motor function of rats after transplantation and made clinical score. I observed rats for four weeks to make spinal cord from all rats into sections, and observed by HE staining method and immunohistochemistry. Another spinal cord was made into ultrathin sectioning and observed by electron microscope.Result:Comparing with the normal saline group, clinical score of treatment group increased. Model group presented myelin swelling and loss of myelin, local inflammatory cell infiltration obviously. Comparing with the normal saline group, myelin swelling eased, the number of inflammatory cells decreased in the treatment group. Comparing with the normal group, the number of myelin which was positive in MBP and Olig2 immunohistochemistry test in model group decreased. Comparing with the normal saline group, the number of myelin which was positive in MBP and Olig2 immunohistochemistry test in model group increased. In the photo of electron microscope, comparing with the normal group, the structure of lamellar myelin was separated. The mitochondria of oligodendroglia swelled. The karyopyknosis was observed. Comparing with the normal saline group, the structure of medullated fibers recovered, the number and structure of organelles in oligodendroglia recovered.Conclusion:The oligodendrocyte precursor cells transplantta-tion had certain effect on repairing the motor function and myelin sheath of the myelinated nerve fibers in spinal cord. |
