[Object] A rapid, accurate and sensitive liquid chromatography-mass spectrometry (LC-MS) method was applied to comprehensively detect and identify the chemical components of Banlangen extract in vitro. On this basis, the absorbed components and their metabolites in rat serum after oral administration of Banlangen extract were identified and characterized according to Serum Pharmacochemistry of Traditional Chinese Medicine(TCM), in order to provide a theoretical basis for further exploring the effective substance of Banlangen.[Method] Combined with Masslynx V4.1 and related literature, the chemical components of Banlangen extract were detected and identified by using a LC-MS method, which was established by the optimization of extraction solvents, chromatographic and mass spectrometric conditions. Then, different serum processing ways were investigated and evaluated to further optimize the analytical method for detecting the components in vivo. At last, the absorbed components and their metabolites in rat serum after oral administration of Banlangen extract were identified and characterized by comparing the chromatograms of Banlangen extract, blank serum and dosed serum at different sampling time.[Results] An injection run of the Banlangen sample was finished in 15 min, and excellent chromatographic separation and sensitive detection were obtained by the established analytical method.40 compounds were identified in Banlangen extract, including amino acids, nucleosides, alkaloids, lignans, et al. The analytical method of the components in vivo was systematically optimized by regarding solid phase extraction as serum processing way, and 32 constituents in vivo were detected in dosed serum at different time. Among them,21 constituents were preliminarily identified and characterized, including 12 prototype constituents derived from alkaloids, organic-acids and 9 metabolites of lignans, flavonoids and alkaloids.[Conclusion] Using the LC-MS technique, the chemical components of Banlangen extract were comprehensively identified, and the results will provide a favorable basis for production process and quality standard study of the Banlangen preparations. Based on Serum Pharmacochemistry of TCM, the absorbed components and their metabolites in rat serum after oral administration of Banlangen extract were identified and characterized for the first time. As a result, epigoitrin, alkaloids, organic acids and lignans were considered as the effective substance of Banlangen.
