Objective To investigate the preservation of glucagon-like peptide-2(GLP-2) on immunologic barrier of intestinal mucosa in rats following small bowel resection.Methods One 30 healthy male rats were randomly assigned into case, control and blank group. The intestinal resection model was established on rats in case and control group. GLP-2 was injected into rats in case group while same concentration saline was injected into control and blank group. The rats were sacrificed at dayO, day 7 and 14 after surgery and the HE staining of mucosa from residual intestine was performed to observe the length, thickness and lacunae of intestinal villus.Methods Two 60 healthy male rats were randomly assigned into no-operation, operation, sham-operation group. And each group was divided into two groups, GLP-2 group and blank group. Same concentration GLP-2 and saline was individually injected into each GLP-2 group and blank group. The rats were sacrificed at day 7 and 14 after surgery and HE staining of mucosa, immunohistochemistry was performed to observe the amount of intraepithelial lymphocyte and slgA expressed by plasma cells located in lamina propria.Results One The value of length, thickness and lacunae of intestinal villus in case group were significantly increased comparing with control and blank groups and significant improvement were also observed in day 14 comparing with day 7 after surgery in case group. Results Two At day 7, the amount of IEL and expression of slgA in GLP-2 group was observed increased than that in blank group either operation or sham-operation group. But there is no statistic difference. At day 14 in operation group, the amount of IEL and the expression of slgA in GLP-2 group increased significantly comparing with blank group (P<0.05); As well the value at day 14 is significantly higher comparing with those at day 7 (P<0.05).Conclusions GLP-2 can effectively improve the proliferation of intestinal mucosa and preserve the mechanical barrier and immunologic barrier of mucosa in rats following small bowel resection and the improvement showed a time-dependent pattern which indicated a fully recover at day 14 after surgery.
