Objective: Observation of digoxin in three negative breast cancer(TNBC) cells and inhibition of MDA-MB-231 nude mice model in vivo and in vitro. Method: Digoxin intervention of breast cancer MDA-MB-231 cells by different concentrations of digoxin after impact detection method of MTT on the proliferation of breast cancer cell MDA-MB-231. The number and morphology of flow cytometry and AO-EB fluorescence staining method for the detection of apoptosis. Expression of Bcl-2 protein and Bax protein was detected in Western Blot. Establishment of MDA-MB-231 cell model in nude mice, to observe the inhibitory effect of digoxin and combined with cisplatin on tumor bearing nude mice model, calculated the tumor inhibitory rate of tumor, the expression of Ki67 in tumor tissues detected by immune group P53. Result: 1. Different concentrations of digoxin in a dose dependent form inhibits proliferation of breast cancer MDA-MB-231 cells. 2. Flow cytometry results show digoxin can significantly induced MDA-MB-231 cells apoptosis, especially late apoptosis. 3. AO-EB fluorescence staining can be obviously observed apoptosis of cell morphology, Western Blot results confirmed that digoxin is by adjusting the bax/Bcl-2 ratio to induce its apoptosis. 4. In vivo experiment results confirm that digoxin has an obvious inhibitory effect on MDA-MB-231 cell xenograft in nude mice, immunohistochemistry method decreased the expression of Ki67 and P53 in transplantation tumor detection within treatment groups. Conclusion: 1. Digoxin can inhibit expression of three negative breast cancer cell MDA-MB-231 proliferation, and down-regulation of nude mice transplanted tumor Ki67, P53. 2. Inhibition of digoxin, cisplatinin human breast cancer xenografts in nude mice tumor growth, enhance the inhibitory effects of combined application of both on the transplantation tumor of breast cancer animal model.
