Objective To study the expression of high molecular weight kininogen(HK)in human clear cell renal cell carcinoma and its clinical evaluation.Method 56 cases of renal cell carcinoma were collected form The First Affiliated Hospital of Soochow University. All the patients with renal cell carcinoma received radical nephrectomy. The expression level of high molecular weight kininogen m RNA in human clear cell renal cell carcinoma.and normal kidney tissues were measurd by real-time reverse transcription polymerase chain reaction(Real-Time PCR) and the expression of HK Protein were measured by immunohistochemistry and western blotting respectively. Furthermore, the relationship between the expression of HK and patients’ clinical material were analyzed with statistical methods.Results(1) The expression level of HK m RNA in human clear cell renal cell carcinoma was higher than normal kidney tissues by using Real-Time PCR method, 21.97±3.03 vs. 7.65±2.96,P<0.05;moreover, the expression of HK protein in human clear cell renal cell carcinoma was higher than normal kidney tissues by using Western blotting method, 0.95±0.12 vs. 0.52±0.13,P<0.05.(2) The expression of HK protein located mainly in cytoplasm of cancer cells.(3) there was no relationship between expression of HK in renal clear cell carcinoma with patients’ age and gender(P>0.05), but there was a significant relationship with histological grade and clinical stage(P<0.05).Conclusion The high expression of HK in human clear cell renal cell carcinoma. It may be play an important role in the development of renal clear cell carcinoma. HK would be one of the potential targets for the treatment in renal carcinoma in the future.
