Objective:The mortality of Esophageal cancer is very high as a common disease, and the 5-year survival rate is only 20%. So it is important to find a kind of drugs with high-efficiency and low-toxicity. Resveratrol in some certain can inhibit a variety of human tumor cells in vitro culture, and it also can produce some cytotoxic. The aim of the study is to investigate the effect of Resveratrol on esophageal cancer and explore the mechanism. And it can provide some theoretical basis for prevention and treatment of esophageal cancer.Methods:EC-9706 cells were exposed to different concentrations of Resveratrol for 24 to 96 hours, respectively.1 MTT assay was used to determine the cell growth inhibitory rate.2 The morphological changes was observed by inverted microscope and transmission electron microscopy.3 Analysis the effect of Resveratrol on EC9706 cell cycle distribution by Flow cytometry.4 Flow Cytometry was used to test the effect of Resveratrol on the expression of EC9706 cells associated protein gene.Results:1 The analysis of MTT:Resveratrol in different concentrations can inhibit EC9706 cell growth. The growth inhibition ratio of EC9706 cells was gradually rising, with the concentration of Resveratrol increasing in some certain time, and it showed the dose-effect relationship. In additional, the growth inhibition ratio was gradually rising, with the different time in a certain concentration, and it showed the time-effect relationship.2 Morphological changes:Under the inverted microscope, we can observed that the particles intracytoplasm enlarged, the cytoplasm became less, the cell became round and small, and the morphology changed, compared with the control group. And by the transmission electron microscope, we can observed nuclear chromatin condensed and focused on the nuclear membrance, density increaesd, the part of the cell membrance to the outside, and the cytoplasm degenerated.3 The analysis of apoptosis and cell cycle was observed by flow Cytometry: After EC9706 cells were empolied by Resveratrol (500μmol/ml, 1000μmol/ml) for 72 hours, the peak of diploid apoptosis occured before G0/G1 phase. The rate of apoptosis was 9.54%±0.52% and 19.85%±0.66%(p<0.01) respectively. The quantity of cells in G0/G1 phase increased, and the percent of cells treated by Resveratrol at 59.5%±1.28% and 68.5%±1.69% respectively, which were higher than the control cells (42.0%±1.23%) (p<0.01). However, the quantity of cells in S and G2/M phase decreased.4 The analysis of the expression of EC9706 cells associated protein gene by flow cytometry:The expression of bcl-2 protein was decreased and the expression of bax protein was increased after EC9706 cells were empolied by Resveratrol (500μmol/ml,1000μmol/ml) for 72 hours.Conclusion:Resveratrol can inhibit the proliferation of esophageal cancer EC9706 cells and induced to apoptosis. So it can block the cell cycle at G0/G1 phase. Also, it can inhibit the cell proliferation by influencing apoptosis-regulated gene bel-2 and bax.
