Objective: to detect the expression changes of mRNAs in pancreas after blockade of the Renin-Angiotensin System(RAS) in Streptozotocin-induced diabetic rats(STZ) treated with Ramipril, and to explore the relation between Reg3α and insular regeneration. Methods: Established STZ induced type 2 diabetic rat model,surveyed the effective in construction and function of pancreatic islet after RAS blocked,investigated the expression pattern of mRNA in pancreas by mRNA array technique, validated by real-time PCR, Western Blotting. Results: Compared with control group rats,the structure of pancreatic islet was be improved; the expression level of 269 mRNAs was deregulated significantly in group rats with Ramipril treatment, 191 of which were up-regulated and the other 78 were down-regulated; Gene expression of Reg3α(regenerating islet-derived 3 alpha) was significantly up-regulated 4.6 multiple, Other islet protection factors such as Nkx6-1, Bcl-2 were also up-regulation 1.6 and 1.7 multiple respectively rats. Witch were validated by real-time PCR, Western Blotting. Conclusion: These findings provided a possible mechanism for the reduced incidence of new-onset diabetes that has been observed in clinical trials of RAS blockade.
