OBJECTIVE A comparative study on the effect of Coffee bean extract on obese rat in two models. To explore the relationship between CBE and weight loss in the model of obese.These exploration will pray the way for the in clinical application of CBE.METHODS The way of rats is based on "security of the State Food and Drug Administration [2012] No. 107" file. To observe the situation of NC、MC and other groups. A record of body weight and food intake is needed before the end of the experiment.The fat weight is also very important. The TC、TG、HDL and LDL are carried out after fasting. The APN and LP are examined by Elisa.The NEFA is detected by testing kit. The expressions of Adipo R2、p38、PGC-1、PPARγ are examined using Western Blot method. The real-time fluorescence quantitative PCR are used to evaluate the Adipo R2、p38、PPARγ、SREBF1、SREBF2 gene expression.RESULTS To establish obesity in male SD rat model of nutrition. Both CBE( 一) and CBE( 二) can reduce the body weight. Specific performance in body weight and fat/weight, there is a significant difference to the normal control group(P<0.01). These two medication groups have significant difference between model control group and normal control group in body weight and fat/weight(P<0.01). The TG and TC value of MC group are significantly higher than the CBE(一) medication groups(P<0.01). The HDL of medication groups are higher than MC. The 133 mg·kg-1、267 mg·kg-1 of LDL have no significant difference between MC group(P>0.05), but the 800 mg·kg-1 of LDL is lower than MC(P<0.05). Compare with NC, the level of APN is lower in MC(P<0.01),the LP increase. The APN of CBE(一) medication groups are significantly higher than MC(P<0.01 或 0.05), but the LP is lower. The expressions of Adipo R2、p38、PGC-1、PPARγ protein on each concentration CBE(一) group has significance in the liver tissues, the expressions of Adipo R2、PGC-1、PPARγ protein in the fat tissues and the expressions of Adipo R2 、 PGC-1 protein in the muscle tissues are significantly higher than MC group, but the expressions of PPARγ can not be detected in the muscle tissues. The results of the real-time fluorescence quantitative PCR showed that the expressions of Adipo R2、p38、PPARγ gene have increased, but the expressions of SREBF1、SREBF2 gene are lower than MC in liver.CONCLUSION The way of rats is based on "security of the State Food and Drug Administration [2012] No. 107" file. Both CBE(一) and CBE(二) have effects on obese rats. Increasing the level of APN and reducing the level of LP may be the major mechanisms, which can promote the expressions of PGC-1 and PPARγ protein, activate the signal transduction on fat tissues and increase the combination of APN and Adipo R2. The downstream signalings of AMPKα、p38、PPARα are activated, and the SREBF1 has dropped off. All above promote lipolysis, inhibit fat production, achieve weight loss.
